Purpose:To observe the effect of TCM-PI Dan Ning on regulating the function of damaged rat islet β cells and its GLUT-2 protein expression.Material and method:In this study,100 male Wistar rats(60-70 days of age,230-250 g in weight)of SPF grade were selected as the experimental subjects,after weight and labeling the experimental rats,the normal control group used a completely random interval method to extract 20 rats.The normal control group was labeled as group A,and the remaining rats experimental mice were used to make a model of impaired glucose regulation.The rats were fed with high-sugar and high-fat diet to damage their glucose metabolism regulation,while the rats in the normal control group were fed with basic diet.After 12 weeks,the rats were tested for glucose tolerance test(OGTT)to confirm the success of the modeling.The matching rats were selected and randomly divided into three groups: The IGR-model control group,the TCM-PI Dan Ning Treatment group,The western medicine metformin treatment group,each group of rats were fed with for four weeks,and fed them separately with 5 ml kg-1 d-1 of distilled water,10 g kg-1 d-1 of TCM-PI Dan Ning decoction,and 150 mg kg-1 d.-1 Metformin suspension;the normal control group was also treated with 5 ml kg-1 d-1 of distilled water.Then,detected and compared the weight of each group of rats before and after,test their fasting blood glucose,OGTT 2 h PG,and HGCT levels to determine the levels of1 Ph and 2Ph secreted by their FINS and INS,and using high glucose clamp technology(HGCT)to measure fasting insulin(FINS),first-phase insulin secretion and second-phase insulin secretion of each group of rats to evaluate islet β-cell function.The rats were sacrificed at the end of the experiment.Fresh rat pancreatic tissue was taken from the abdomen,and the expression of GLUT-2 protein on pancreatic β-cells was observed by Western blotting.Results:1.Islet β-cell secretion function: Before drug intervention,insulin secretion at the first and second phases of the model control group,the TCM-PI Dan Ning treatment group,and the metformin treatment group was significantly lower than the normal control group(P <0.01).There was no statistical difference between the groups(P> 0.05).After the drug intervention,the levels of insulin secretion in the first and second phases of the TCM-PI Dan Ning treatment group were significantly lower than those in the normal control group(P <0.01),but higher than those in the model control group(P <0.05).The levels of insulin secretion in the first and second phases of the metformin treatment group were lower than those in the normal control group(P1 <0.01,P2 <0.05),and significantly higher than those in the model control group(P <0.01).The second phase of insulin secretion in the metformin treatment group It was significantly higher than that of the TCM-PI Dan Ning treatment group(P <0.05).2.GLUT-2 protein expression on pancreatic islet β cells: The expression of GLUT-2 protein on pancreatic islet β cells in the rats of the TCM-PI Dan Ning treatment group,and metformin treatment group was significantly up-regulated compared with the model control group,but still failed to reach the normal control The state of the group(P <0.01);the ability of the TCM-PI Dan Ning treatment group,to upregulate the expression of GLUT-2 protein in islet βcells was more prominent than that of metformin(P <0.05).Conclusion:1.TCM-PI Dan Ning can significantly improve the function of pancreatic islet β cells in rats with impaired glucose regulation.2.TCM-PI Dan Ning can significantly increase the amount of GLUT-2 protein on pancreaticβ-cells in rats with impaired glucose regulation.3.TCM-PI Dan Ning can significantly increase the secretion function of islet cells by increasing the glucose regulation of GLUT-2 protein expression on β-cells of islet model rats.
