Purpose:Study on Chinese herbal compound Pidan Ning on rat model of impaired glucose regulation to the function of pancreatic beta cells and the expression of free fatty acid.Material and method:A total of 100 male Wistar rats aged 60-70 days and SPF grade were randomly divided into normal control group(n=20)and impaired glucose regulation group(n=80)according to body weight.They were fed basal diet and high glucose and high-fat diet respectively,and 12 weeks later,the oral glucose tolerance test(OGTT)was performed.Impaired glucose regulation after the success of modeling,will reach the impaired glucose regulation model rats were randomly divided into model group,Pidan Ning treatment group and metformin group were gavage for 4 weeks.Before and after treatment were measured the weight of rats,fasting blood glucose(FBG)and fasting insulin(FINS),to evaluate the function of islet beta cells by high glucose clamp technique.abdominal aortic blood test,the content of free fatty acid(FFA)was measured by copper reagent staining.and analysis the correlation between of FFA and pancreatic beta cell secretory function of island.At the end of the experiment,pancreatic tissue was removed by laparotomy,and the apoptosis of islet cells was assessed by in situ terminal labeling(TUNEL).The correlation between FFA and islet cell apoptosis was analyzed.Results:1.beta cell function: before treatment model control group,Pidan Ning treatment group and metformin treatment group in the first phase and the second phase of insulin secretion was significantly lower than normal control group(P<0.01),no significant differences between the three groups(P>0.05).After medication Pidan Ning treatment group first phase insulin secretion and second phase levels were significantly lower than the normal control group(P<0.01),significantly higher than the model control group(P<0.05).The metformin group first phase insulin secretion and second phase levels were significantly lower than normal control group(P<0.01,P<0.05),were significantly higher than the model group(P<0.01);metformin treatment group the second phase of insulin secretion was significantly higher than that in Pidan Ning treatment group(P<0.05).2.FFA level: before treatment model control group,Pidan Ning treatment group and metformin treatment group FFA was significantly higher than the normal control group (P<0.01),no significant difference between the three groups(P>0.05).After medication Pidan Ning treatment group and metformin treatment group FFA was significantly higher than the normal control group(P<0.01),significantly lower than the model control group(P<0.01).3.the level of islet cell apoptosis: after drug intervention Pidan Ning treatment group and metformin treatment group was significantly higher than the number of islet cell apoptosis in normal control group(P<0.01),significantly lower than that of model group(P<0.01).4.the correlation among FFA and first phase and the second phase of insulin secretion,islet cell apoptosis: correlation between FFA and the first phase and the second phase of insulin secretion was negatively correlated(r=-0.734,P<0.01,r=-0.722,P<0.01);FFA was positively correlated with the apoptosis of islet cells(r= 0.857,P<0.01).Conclusion:1.Pidan Ning can significantly improve the islet beta cell function of rat model of impaired glucose regulation.2.Pidan Ning can significantly reduce the concentration of FFA,for the rat model of impaired glucose regulation.3.Pidan Ning can significantly increase the islet cell secretion,for the rat model of impaired glucose regulation.4.Pidan Ning can significantly reduce the apoptosis of islet cells,for the rat model of impaired glucose regulation.5.Pidan Ning can reduce the concentration of FFA concentration for rat model of impaired glucose regulation,to enhanced insulin secretion and reduce the apoptosis of islet cells to improve the function of beta cell. |
