Based On The HSP70/NF-κB Pathway, Jianpi Qinghua Decoction Treats The Mechanism Of Hp-related Gastritis In Rats

Purpose:To observe the general effects of H.pylori-associated gastritis rats before and after intervention with different concentrations of Jianpi Qinghua Decoction,13C urea breath test,and pathological changes of gastric mucosal epithelium in mouse models,and to explore the expression of HSP70 and NF-κBp65 Possible mechanism of Jianpi Qinghua Decoction in the treatment of Hp-related gastritis.Methods:(1)Experimental process:66 healthy male SD rats were randomly divided into 6 groups randomly,blank group,model group,western medicine teprenone group,Jianpi Qinghua Decoction high-dose Chinese medicine group,Jianpiqing Huatang medium-dose Chinese medicine group,Jianpi Qinghua decoction low-dose Chinese medicine group.Except the blank group,the other 5 groups were administered with Hp bacterial solution(109CFU/ml)by gavage to establish an Hp gastritis model.After 5 times of continuous bacterial solution intragastric administration,the change of BOD value was detected by 13C breath test detection method.After successful modeling,tepredone and the corresponding concentration of Jianpi Qinghua Decoction were administered intragastrically for one month,and samples were taken after the treatment was completed.(2)Detection indicators:HE staining under light microscope was used to observe the histopathological changes of gastric mucosa,and immunohistochemical staining was used to detect the expression of HSP70 and NF-κBp65 protein in gastric mucosal epithelial cells.Results:(1)The general conditions of rats in the high,medium and low dose of Jianpi Qinghua Decoction group and the western medicine group were improved.(2)13C breath test:The DOB value of rats after modeling was significantly higher than the DOB levels before modeling and the blank group,and the difference was statistically significant(P<0.05).One month after the administration,the model group was statistically significant compared with the western medicine group and each traditional Chinese medicine group(P<0.05),and the average DOB value of the western medicine group and the traditional Chinese medicine group decreased.The middle and high dose groups were statistically significant(P<0.05).(2)HE staining:the model group had more damage than the blank group;the western medicine group and the middle-dose Chinese medicine group had better damage than the model group.The number of intact gastric mucosal epithelial cells in the model group was lower than that in the normal group;the number of intact cells in the western medicine group and the high and medium dose Chinese medicine group was higher than the model group;(3)NF-κBp65 immunohistochemical staining:NF-κBp65 in the gastric mucosa of rats in the blank group Negative or very little positive expression,mainly expressed in the cytcplasm;Compared with the blank group,the model group NF-κB65 was expressed in the cytoplasm and nucleus,showing dark brown,scattered in the cytoplasm and nucleus,the protein expression level was significantly increased,the average The highest optical density value;NF-κB65 in the western medicine group and Jianpi Qinghua Decoction high and medium dose groups are expressed in the cytoplasm and nucleus,and a small part is expressed in the nucleus.The average optical density is lower in the Chinese medicine group and the low dose in the traditional Chinese medicine group.NF-κB is mainly expressed in the cytoplasm,and a small part is expressed in the nucleus,which is slightly lower than the model group;the average optical density of positive cells in the low,high,and medium dose groups of Jianpi Qinghua Decoction decreased in order.In Jianpi Qinghua Decoction,the average optical density of positive cells in the low,high and medium dose groups decreased in turn.Western medicine group and Jianpi Qinghua Tang high-dose group and middle-dose group AOD values were lower than the model group;Jianpi Qinghua Tang low-dose group was slightly lower than the model group.HSP70 immunohistochemical staining:a small amount of positive expression of HSP70 in the gastric mucosa of rats in the blank group,mainly expressed in the cytoplasm,rare and nucleus,showing light brown;compared with the blank group,the model group HSP70 expression is very small,rarely seen in scattered In the cytoplasm,the average optical density value was the lowest in each group;the Western medicine teprenone group showed a strong positive expression of HSP70,distributed in the cytoplasm and nucleus,showing a dark brown color,the protein expression level was significantly increased,and the average optical density value was high;Jianpi HSP70 in the medium-dose group of Qinghua Decoction is mainly expressed in the cytoplasm and nucleus,and the average optical density value is not significantly different from that in the western medicine group;HSP70 in the high-dose and low-dose groups of Jianpi Qinghua Decoction is mainly expressed in the cytoplasm,and a small part In the nucleus,it is brown,and the average optical density is not significantly changed compared with the blank group;the average optical density of the positive cells in Jianpi Qinghua Decoction group from high to low is the middle dose of Jianpi Qinghua Decoction in order.Group,high dose group,low dose group.The AOD value of the model group was lower than that of the blank group;the AOD values of the western medicine group and Jianpi Qinghua Decoction were higher than that of the model groupConclusion:Jianpi Qinghua Decoction can inhibit Hp to a certain extent while improving the general condition of SD rats.HSP70,which was originally suppressed due to Hp infection,can be restored under the action of Jianpi Qinghua Decoction.The expression of HSP70 makes the expression of NF-κB It is suppressed,thereby reducing the progress of the inflammatory response,reducing gastric tissue damage to protect the stomach tissue.
