Perfluorinated compounds have garnered enormous interest for their high oxygen loading.Currently they have been applied in adjuvant treatment for tumor,ultrasound imaging,organ perfusion and ophthalmic surgery.Nevertheless,researches have shown that perfluorinated compounds cause many side effects including thrombocytopenia,flu-like symptoms,multiple organ pain and stroke,etc.These side effects rely on emulsifiers like poloxamer,and the category,amount and distribution in vivo of perfluorinated compounds.To reduce the side effects of perfluorinated compounds in vivo,in this paper,two kinds of perfluorinated compounds are used to prepare a novel liposome of perfluorinated compounds(PFC@Lip).The preliminary investigation and evaluation of the shape,size and storage stability of perfluorinated compounds are performed.The oxygen carrying ability is also examined using hypoxic cells model.In addition,the acute toxicity of PFC@Lip are examined both in vitro and in vivo.Lastly,a gas chromatography method has been established for accurate measurement of the amount of perfluorinated compounds in PFC@Lip through optimization of uncertain parameters.The results show that PFC@Lip is safe and capable of reverting the hypoxic state of cells in vitro.Moreover,the gas chromatography method we established is promising to be used for the quantitative analysis of other perfluorinated compounds.Otherwise,we also use transferrin and IR780,which is a photosensitizer of near-infrared small molecules,to preparate a nanoparticle called IR780@Tf.Then,we show that the nanoparticles have the double therapy of photothermal and photodynamic in aqueous solution and cellular level in vitro respectively.
