In recent years,natural active substances have attracted a lot of attentions and developing new multifunctional natural active substances become a hot topic.Flavonoids are new nature active substances contained in bamboo leaves with the effect of multiple physiological,which are worth to explore.There are many bamboo resources in Chishui City in Guizhou province,but there is still no relevant reports on the study of the deep processing of bamboo leaves so far and the waste of bamboo resources is still serious.This paper provide reliable data for the industrialized production of bamboo leaf flavone and determine the best pilot extraction process parameters based on laboratory scale test results.The optimal pilot parameter was determined by compared the bamboo leaf flavonoids rate,extraction rate in different conditions of different solid-liquid ratio,extraction time,extraction temperature and the concentration of alcohol.The best process for the production of flavonoids as follow: first soaked 0.5h,then two times extraction in the solid-liquid ratio of 1:15(kg/L),alcohol concentration 60%,extraction temperature 80 ℃ and extraction time is 1.5h and 1h.The optimum extraction process experiments show that the pilot tests process is basically stable and feasible,the purity of flavonoids was 2.0~2.15%,extracting rate was 74~80%.There are many impurities in the bamboo leaf extract by the pilot test,in order to obtain a higher purity of bamboo leaf flavonoids,so that it can be applied to the high demand of health products and food industry.By comparing the two types of resins for purify the bamboo leaf flavonoids,studying on thermal stability of the bamboo leaf flavonoids and resin’s regeneration process.indicate that the purification effect of AB-8 resin is better than the D101 resin,the purity of sample was 23%~26% and the process is basically stable and feasible.Furthermore,loss rate of purified bamboo leaf flavonoids in the process of heat recovered solvents is lower than without purification.Macroporous resin adsorption rate decreases along with the two short-term regeneration,which needs to go through strengthening regeneration before use.HPLC method was used for quantitative analysis of homoorientin,orientin,isovitexin and vitexin after purification in bamboo leaf flavonoids.The chromatography conditions were described as follows: with A,B two pump systems,mobile phase A(methanol): B(0.3% aqueous acetic acid,)=32:68(v/v)、 flow rate of mobile phase 1 m L/min、the column temperature was 20 ℃;the injection volume was 5μL and detection wavelength was 340 nm.The results shows that four kinds of flavone glycosides cold be separated within 30 min.The research on HPLC detection method of flavonoids in bamboo leaf was evaluated from precision test and stability.This method is rapid and accurate,so it can be applied to determination bamboo leaves flavonoid C-glycosides content.The purity of flavonoids do not meet the requirements of dietary supplements(≥24%~32%)after pilot purification process.This subject aim to obtain higher purity of bamboo leaf flavone by using macroporous resin mixed technology.The best conditions of mixed resin purification bamboo leaves was sifted from eight kinds of macroporous resin and a mixed resin static adsorption-desorption experiments and dynamic experiments.The experiments show that the ratio of D101-1 and AB-8 is 2:1,the sample conditions:the sample concentration is 1.2 mg/m L,p H is 3,sample volume is 3.5BV,the flow rate is 2BV/h.elution conditions:eluent volume fraction of 70%,the eluent volume 4BV,eluent flow rate 2BV/h.After the process,the purity of flavonoids becomes tripled from 8.46% to 28.14%.It is clear to see that purification effect is remarkable and offer a new method for purification bamboo leaves.The bamboo leaves in Chishui city of Guizhou province are rich in flavonoids which are multiple uses such as feed additives,dietary supplements after purification.It is promising to take full advantage of it.This subject implements the bamboo leaves extracted from laboratory scale to large-scale production of transition and provide reference for the development of the region bamboo leaves flavoniods industrialization.
