Studies On The Chemical Constituents And Bioactivity Of Hedyotis Uncinella

“Dui Zuo Ye”,Hedyotis uncinella Hook.et Arn,is a plant from the genus Hedyotis L.which belongs to the family Rubiaceae.H.uncinella is mainly distributed in Hainan,Guangdong,Hunan,Guizhou,Taiwan and Hong Kong provinces,and grows on sparse forests and dry land.H.uncinella is minority medicine in Guizhou,it is called “Xia Jia Liu”.H.uncinella has long been used as a common folk medicine in Guizhou for the treatment of rheumatism,diarrhea,dysentery,hepatitis,and snakebite.The extract of H.uncinella has been used to produce the traditional Chinese medicines.This research focuses on the study of chemical constituents and pharmacological activities of the whole plant,providing theoretical basis for further studies on the H.uncinella.Application of various chromatography materials(such as silica gel,MCI,Sephadex LH-20)and separation methods(column chromatography,TLC and HPLC),a total of 26 compounds were isolated from the plant and identified according to modern spectroscopy techniques,physical and chemical properties: 3-formyl-oleanolic acid(DZY-1),3β-hydroxy-11-oxo-oleana-12-en-28-oic acid(DZY-2),2α,3β-dihydroxy-olean-12-en-28-oic acid(DZY-3),oleanolic acid(DZY-4),ursolic acid(DZY-5),3β,19α-dihydroxy-urs-12-en--28-oic acid(DZY-6),sorghumol(DZY-7),19,28-didehydroxy-rubiarbonol A(DZY-8),3β-hydroxylup-12-en-28-oic acid(DZY-9),β-sitosterol(DZY-10),7β-hydroxystigmasterol(DZY-11),scopoletin(DZY-12),lapachol(DZY-13),3-hydroxy-4-methoxycinnamaladehyde(DZY-14),1,1’-oxybis-2,5-bis-(1,1-di-me-thylethyl)-benzene(DZY-15),benzoic acid(DZY-16),loliolide(DZY-17),(6R,9S)-3-Oxo-α-ionol(DZY-18),cassipourol(DZY-19),(2S,6S)-2,6-dimethyl-7-octene-1,6-diol(DZY-20),myristic acid(DZY-21),α-linolenic acid(DZY-22),n-octacosan-7β-ol(DZY-23),phytol(DZY-24),palmitatic acid(DZY-25),octacosanol(DZY-26).DZY-1 is a new compound,and DZY-2,DZY-3,DZY-7,DZY-8 and DZY-9 were isolated from the genus Hedyotis L.for the first time,and with the exception of DZY-4,DZY-5,DZY-10 and DZY-12,22 compounds were isolated from H.uncinella for the first time.The type of these compounds include triterpenoids,steroids,quinones,coumarins,aliphatic and others.The antioxidant activity of some isolated compounds was evaluated by DPPH free radical scavenging experiment and superoxide anion free radical scavenging experiment.In addition,some compounds were evaluated for anti-tumor activity in vitro by the MTT method.The experimental results found that the compound DZY-6 has the best inhibitory effect on the three tumor cell lines and has a good selectivity index for Hep G2 cancer cells.
