| Objective: Genetic factors play an important role in the pathogenesis of osteoporosis(OP).Vitamin D receptor(VDR)genes regulate calcium uptake,bone resorption,osteocyte differentiation and parathyroid hormone secretion.The purpose of this experiment is to explore the relationship between OP and VDR gene polymorphism in the Han and She populations in Putian area.Methods: From September 2018 to October 2020,Han and She OP patients in Putian area were recruited in the general practice department and geriatrics ward of our hospital in the same age group for the study,and the healthy population was used as the control group.There were 114 cases in Han population(37 OP patients,77 healthy control group)and 188 cases in She population(53 OP patients,135 healthy control group).Clinical characteristics such as age,weight,height,body mass index and bone mineral density were collected.After blood samples were taken in the morning,calcium,phosphorus,vitamin D3,osteocalcin,estradiol,testosterone,parathyroidin,total Ⅰcollagen amino-terminal-elongation peptide and alkaline phosphatase were measured.VDR gene polymorphism was analyzed after enzyme digestion with ApaI,TaqI,BsmI and FokI,respectively.Compare the difference of data between Han and She OP patients and healthy control group in Putian area.Results: There were no significant differences in the average age,height,weight,body mass index and bone mineral density between the two OP groups(P>0.05).The total Ⅰ amino-terminal elongation peptide of type collagen in the She healthy control group was significantly lower than that in the Han healthy control group(P<0.05).Compared with OP group,there were significant differences in age,height,weight,body mass index,osteocalcin,total Ⅰ amino prolongation peptide and bone mineral density in Han and She healthy control group(P<0.05).There were significant differences in the FokI zymotypes of VDR gene between healthy population and OP population(P<0.05),but there were no significant differences in the other three zymotypes.Compared with the healthy control group,the Ff genotype frequency of She OP patients was significantly higher and the ff genotype frequency was significantly lower,while the ff genotype frequency of Han OP patients was significantly higher(P <0.05).The frequency of bbaa TTFf combination genotype was the highest in OP patients in She and Han populations.There was no significant difference in bone mineral density between the Han and She OP genotypes(P>0.05).There were significant differences in alkaline phosphatase among BsmI genotypes and phosphorus among TaqI genotypes in She population(P<0.05).There were significant differences in vitamin D3 among ApaI genotypes in Han population(P<0.05).Conclusions: There is no significant correlation between BsmI,ApaI and TaqI restriction sites polymorphisms and OP susceptibility in Han and She populations in Putian area.FokI gene polymorphism has an important relationship with the development of OP in the Han and She populations in Putian.Ff genotype is a susceptibility gene for She people,while ff genotype is a protective factor.The genotype of ff is a susceptibility gene in OP population of Han nationality.The relationship between VDR genotype polymorphism and various bone biochemical indexes and bone mineral density in OP patients is not obvious.Therefore,large sample studies of different populations are needed to confirm the influence of genetic factors on OP patients. |
