| Objectives It is intended to analyze the toxicity source of compatibility of Erxian couplet medicines and to study the correlation between their toxicity and the content of medicines with a series of compatible prescription;to explore the optimal compatibility of Erxian couplet medicines in improving effects and decreasing toxicity;to research the invigorating the kidney and strengthening Yang function of compatibility of Erxian couplet medicines and its possible pharmacological mechanism.Methods(1)The toxicity measurement of compatibility of Erxian couplet medicines and the correlation research of toxicity and contentUnder the premise of constant quantity,with the classical proportion of curculigo rhizome and epimedium 1:1 as the benchmark,it utilized the design method of baseline geometric changes to develop the right ratio of curculigo and epimedium drugs and prepare single curculigo rhizome,epimedium pubescens as well as extracts of their compatibility prescription;it made comparisons of the acute toxicity of mice in each group;then,the contents of curculigoside and monosomic icariin in each compatibility prescription were determined by HPLC method for analyzing the correlation between toxicity and content.(2)The comparison research of the efficacy results of Erxian couplet medicines compatibility between two kinds of kidney-yang deficient miceBased on establishing models of mice with hydrocortisone and adenine induced kidney-yang deficiency,it observed and compared the effects of single curculigo,single epimedium and its compatibilities for invigorating kidney and strengthening yang of those kidney-yang deficient mice.Then,the best compatibility of epimedium and curculigo to improve effect and decrease toxicity was preliminarily screened.(3)The research of efficacy Erxian couplet medicines compatibility on kidney-yang deficient ratWith the symptoms of kidney yang deficiency,ACTH,IL-2 and BGP as the indicators,it observed and compared the effects of single curculigo,single epimedium and its compatibilities for invigorating kidney and strengthening yang of those kidney-yang deficient mice so as to determine the best compatibility.Results(1)The toxicity measurement of compatibility of Erxian couplet medicines and the correlation research of toxicity and contentThe acute toxicity sequence was alcohol extract of curculigo-epimedium(1:0)>alcohol extract of curculigo-epimedium(2:1)>alcohol extract of curculigo-epimedium(1:1)>alcohol extract of curculigo-epimedium(1:2)>alcohol extract of curculigo-epimedium(0:1)>water extract group.There was significantly positive correlation between the content of curculigoside and its toxicity;No correlation was found between the content of Icariin and its toxicity.(2)The comparison research of the efficacy results of Erxian couplet medicines compatibility between two kinds of kidney-yang deficient mice.The efficacy results of models of mice with hydrocortisone induced kidney-yang deficiency show that:①mice of water extract 1:1 and 1:2 groups,alcohol extract of 2:1 and 1:2groups and alcohol extraction Epimedium group,a significant increase in body weight of mice;②water extract 1:1 and 1:2 groups,alcohol extract of 1:1,2:1 and 1:2groups and alcohol extraction Epimedium group,time of low-temperature swimming for mice increase is significant;③water extract curculigo group,water extraction 1:1 and 1:2 groups,viscera index increase was most pronounced;④water 1:2 group significantly increased serum SOD activity in mice;water extract 2:1 and 1:2 groups,water extract Curculigo group,alcohol extract 1:2 group and the alcohol extract epimedium group significantly reduce serum MDA content in mice.For all the efficacy results of models of mice with hydrocortisone induced kidney-yang deficiency show that,Of all the compatibilities,water extraction or alcohol extraction of curculigo-epimedium(1:2)has the most distinct effects in invigorating kidney and strengthening yang of those two kinds of mice.The efficacy results of models of mice with adenine induced kidney-yang deficiency show that:① After three weeks experiment,the mice of water extract 1:1 and 1:2 group,water extraction Epimedium group and alcohol extract of I:2 and 1:1 groups,a significant increase in body weight of mice;② water extraction 1:2 groups and alcohol extract 1:2 and 1:1 groups of independent activity is very significant increase in the number of mice;③water Epimedium group and alcohol extract 2:1 and 1:1 groups,viscera index increased more obviously;④ water extraction 2:land 1:2 groups,and alcohol extract 1:2 group of mice were significantly increased plasma cAMP vitality,ACTH were significantly increased.For all the efficacy results of models of mice with adenine induced kidney-yang deficiency show that,Of all the compatibilities,water extraction or alcohol extraction of curculigoepimedium(1:2)has the most distinct effects in invigorating kidney and strengthening yang of those two kinds of mice.(3)The research of efficacy Erxian couplet medicines compatibility on kidney-yang deficient ratCompared with normal control group rats,the weight and water quantity of model group rats was declined,but no obvious change in food intake;The levels of BGP、ACTH and IL-2 were significantly reduced;On the contrary,Viscera weight and viscera index were increased.After dosing four weeks,compared with model group rats,the weight of each dosage groups had a certain degree of increase,among them,water extraction 1:0 group has significant differences;compared with model group rats,the food intake of each dosage groups had no obvious change,but the water quantity had a certain degree;compared with model group rats,groups of serum BGP were all increase and the alcohol extract 1:2 group has significant differences;compared with model group rats,groups of serum ACTH were all increase and the water extract 1:0 and 1:2 groups and alcohol extract 1:2 group had significant differences;compared with model group rats,groups of serum IL-2 were all increase and the water extract 1:0 group and alcohol extract 1:2 group had significant differences.Groups of various viscera weight and viscera index has a certain difference,but the directivity is not unified,compared with model group,all dosage groups had a certain improvement effect.Conclusion(1)The compatibility of curculigo-epimedium contains the functions of enhancing effects and reducing toxicity,and their toxic components mainly come from curculigo and they are also easily enriched in alcohol extracts.(2)Of all the compatibilities,curculigo-epimedium(1:2)has the most significant effect in invigorating the kidney and strengthening Yang,and it is a good compatibility to enhance efficiency and reduce toxicity.(3)Erxian couplet medicines compatibility not only enhances the function of invigorating the kidney and strengthening yang,but also significantly improves the contents of BGP、ACTH and IL-2 of the animal model with Yang deficiency syndrome. |
