Part ?:The effects of tortoise deer erxianjiao on the antioxidant and pathological damage of testicular tissues in rats with kidney Yang deficiency and hypoasthenospermiaObjective: to study the effect of tortoise deer er xian jiao on antioxidant and pathological damage of testicular tissue in rats with kidney Yang deficiency and oligospermia.Methods: 40 male SD rats were randomly divided into normal group(10)and model group(30).After the model was successfully constructed with 200mg/kg adenine suspension,the model group was randomly divided into the model group,the levocarnitine group,the kirin pill group,the tortoise deer er xian jiao high-dose group,and the low-dose group,with 5 mice in each group.The corresponding drugs were given continuous gavage,respectively,and the normal group and the model group were given normal saline gavage,4m L/d.The rats were anesthetized 4 weeks later.The epididymis was isolated and extracted from the rats.The epididymis was cut into pieces in a physiological salt level dish,mixed and bathed in a warm bath(around 30s).Sperm count and sperm motility were measured by sperm counting board and CASA after the sperm had fully spread.The contents of SOD,MDA and GSH-Px in the homogenized testicular tissue on one side were detected by ELISA,and the testicular tissue on the other side was analyzed by routine pathological section.Results: Compared with model group,the blank group,high dose group of SOD,GSH-Px concentration significantly increased(P <0.01),MDA concentration decreased significantly(P < 0.01),low dose group of SOD has no obvious change no statistical significance(P > 0.05),low dose group,l-carnitine,GSH-Px concentration increased significantly(P < 0.01),MDA concentration decreased significantly(P < 0.05),l-carnitine group of SOD increased significantly(P < 0.01),kirin pill group significantly increased concentrations of SOD,GSH-Px(P < 0.01),MDA concentration decreased significantly(P<0.01),which was statistically significant.Compared with the high-dose group,the MDA concentration in the low-dose group was significantly increased(P<0.01),and the concentrations of SOD and GSH-Px were significantly decreased(P<0.05).The difference was statistically significant.Compared with the low-dose group,there was no significant change in the indicators of l-carnitine group and kirin pill group(P>0.05),showing no statistical significance.The results showed that tortoise deer er xian jiao could enhance the activity of antioxidant enzyme in rat testis,and had the effect of improving the antioxidant of testis tissue.Compared with model group,the blank group,high dose group,l-carnitine,sperm density,vitality,living rate,activity significantly increased the total number of sperm(P < 0.01),kirin pill group of sperm density,vigor significantly increased(P < 0.01),energy,total number of sperm activity significantly increased(P < 0.05),low dose group,the total number of sperm,sperm density,living rate increased significantly(P < 0.05),energy increased significantly(P < 0.01),there is statistical significance;Compared with the high-dose group,the low-dose group,kirin pill group,and l-carnitine group had no significant changes in sperm density,motility,viability,and total number of motile sperm(P>0.05),which were not statistically significant.In the model group,the epithelium of the testicular convoluted fine tubules was significantly thinner,some of the tubules were wider,spermatogonial cells at all levels in the lumen were rare,the cell arrangement was loose and disordered,and the structure of the spermatogenic tubules was fuzzy.Testicular spermatogenic dysfunction,sperm cell number significantly reduced.The testicular histopathological changes were similar in the groups of levocarnitine,kugelu erxianjiao and qilin pill.Spermatocyte cells were found at the edge of the convoluted fine semen tube in testicular tissue,but the arrangement was irregular.The number of sperm cells in the lumen decreased obviously compared with the normal.The testicular histopathological changes of rats in the high-dose group showed that the epithelium of spermatogenic tubules was still intact.Although the arrangement of spermatocytes at all levels of the tubules was slightly disordered,a large number of spermatocytes were found to have formed in the tubules,and the tissue morphology and structure were normal.The results showed that tortoise deer erxianjiao could significantly improve the sperm quality of rats with kidney Yang deficiency and oligospermia.Conclusion: tortoise deer er xian jiao may improve the antioxidant and pathological damage repair of testicular tissue in rats with kidney-yang deficiency and oligoasthenospermia,and improve the sperm quality of rats.Part ?: Effects of tortoise deer er xian jiao on the expression of Bcl-2 and Cyt-c proteins in testicular tissues of rats with kidney Yang deficiency and oligospermiaObjective: to study the effect of tortoise deer er xian jiao on the expression of Bcl-2 and Cyt-c in testicular tissues of rats with kidney Yang deficiency and oligospermia.Methods: modeling,grouping and drug administration were the same as experiment.After 28 days of administration,the rats were anesthetized with 10% chloral hydrate(3m L/kg).One side of testicular tissue was taken and the expression levels of Bcl-2 and Cyt-c in testicular tissue were determined by immunohistochemical Mean Density analysis.Results: compared with the model group,the positive expression of Bcl-2 in blank group,l-carnitine group,Kirin pill group,low-dose group and high-dose group was significantly increased(P<0.01),and the positive expression of Cyt-c protein was significantly decreased(P<0.01).Compared with the high-dose group,the positive expression of Bcl-2 protein in the low-dose group was significantly reduced(P<0.01),and the positive expression of Cyt-c protein was significantly increased(P<0.01).The differences were statistically significant.Compared with the low-dose group,there was no significant change in the indicators of l-carnitine group and Kirin pill group(P>0.05),and the difference was not statistically significant.Conclusion: It may be one of the mechanisms for the treatment of oligoasthenospermia to regulate the expression of Bcl-2 and Cyt-c in rat testicular tissues,regulate the apoptosis of spermatogenic cells,and improve the spermatogenic function.
