| Capparis spinosa L.(Capparis spinosa L.)is a plant of the genus Capparis in the Capparineae family.Its fruits are rich in sugars,alkaloids,flavones,citrus,fatty acids,volatile oils and steroids.It is used by the people to control obesity,High blood pressure,arthritis,rheumatism,kidney disease and other diseases,according to literature reports,water extract of Capparis spinosa L.has a hypoglycemic effect,but the chemical components and mechanisms of the hypoglycemic activity of Capparis spinosa L.fruits are unknown.Numerous studies have shown that polysaccharides have unique hypoglycemic effects.This paper mainly studies the optimization of the Capparis spinosa L.crude polysaccharides preparation process.The obtained crude polysaccharides are used to intervene in the diet of obese diabetic mice to conduct preliminary research on the mechanism of hypoglycemia,as well as to consider the application of reserves and other literature analysis The crude polysaccharides was prepared into granules,and its preparation process was investigated.Therefore,it provides a research basis for the further development and utilization of Capparis spinosa L.polysaccharides.Crude polysaccharides of Capparis spinosa L.were extracted from the dried fruits of Capparis spinosa L.by hot water extraction method.The polysaccharides content was determined by phenol-sulfuric acid method combined with ultraviolet spectrophotometry.Taking ethanol concentration and pigment as influencing factors and crude polysaccharides content as evaluation index,the preparation technology of crude polysaccharides from Capparis spinosa L.was optimized by orthogonal experimental design.The results show that the optimal purification process of polysaccharides is that the concentration of alcohol is 90 %,freeze-dried into powder after dialysis,the content of crude polysaccharides is diluted to colorless when determining the content of crude polysaccharides,and the content of crude polysaccharides is 94 %.It is concluded that the preparation process is stable and reliable,has low equipment requirements and is easy to control.It can be considered for production use.In vivo experiment,the mice were fed with 45 % high-fat diet(HFD)for several weeks to establish the model of obese diabetes.In the treatment group,crude polysaccharides from Capparis spinosa L.were added to the diet of mice fed with high-fat diet for 12 weeks,and the body weight was recorded every week.The glucose tolerance test(IPGTT)was performed on the 10 th week and the insulin tolerance test(IPITT)was performed on the 11 th week.The results showed that the intervention of Capparis spinosa L.crude polysaccharides on the diet of obese mice could reduce their body weight.IPGTT results show that Capparis spinosa L.polysaccharides can reduce fasting blood glucose in obese mice,significantly increase glucose tolerance and repair impaired glucose tolerance in diabetic mice;IPITT experiments have verified that Capparis spinosa L.polysaccharides can improve its insulin sensitivity.Finally,it is concluded that Capparis spinosa L.polysaccharides has good hypoglycemic activity,which can improve impaired glucose tolerance and insulin resistance in obese mice,control the return of blood glucose levels to the normal range,and can be deeply developed and used reasonably.Finally,the preparation process of Capparis spinosa L.crude polysaccharides granules was preliminarily studied,and the granulation process was screened by single factor test.Finally,the best prescription process was determined as follows: the crude polysaccharides powder was mixed with the excipient dextrin at 1:5,and 80 % ethanol was used as the wetting agent.The three batches of granules prepared according to the above-mentioned prescription process all meet the standards of the 2015 edition of Chinese Pharmacopoeia.In this paper,the quality standard of Capparis spinosa L.granules was studied and the experimental results show that the preparation process is good and the stability of the granules is good. |
