Research On The Law Of Traditional Chinese Medicine Prescriptions For Lower Deep Vein Thrombosis Based On Traditional Chinese Medicine Inheritance Support System

Objective:The clinical treatment of traditional Chinese medicine(TCM) plays an important role in the process of protecting people’s life and health,and prescription is the main form of clinical application of TCM.It is an important content of inheriting the methods of TCM disease prevention and treatment to study and summarize the rules of prescription medication.Lower deep vein thrombosis(DVT)is a common clinical peripheral vascular disease.However,there are few literatures on the drug prescription and mechanism of action of DVT.Therefore,based on the Traditional Chinese Medicine Inheritance Support System(TCMISS),this study summarized the medication rules of lower DVT and speculated drug-disease targets and related pathways by using network pharmacology technology.Methods:The literature of clinical randomized controlled trials(RCTs)with complete observation indexes and efficacy evaluation indexes in the treatment of lower DVT by TCM from January 1990 to December 2020 in four databases including CNKI was retrieved by combining keywords with subject words.According to the inclusion criteria,exclusion criteria and diagnostic criteria of the research literature,the target literature was screened and obtained.Then,the article titles,data sources,prescription names,Chinese medicine prescriptions and doses in the literature were input into Excle to develop a literature questionnaire on the lower DVT.Before the study of data entry into the TCMISS,the name and dosage of TCM prescriptions should be unified and standardized.The statistical report and data analysis function of the platform were used to conduct statistics on the drug frequency,four qi and five flavors,and return meridian of the included prescriptions,and then association rule analysis and cluster analysis were used to obtain high-frequency drug combinations and new prescriptions.Finally,through the traditional Chinese medicine system pharmacological database and other platforms,the main active compounds of high-frequency drugs were screened and target prediction was made,and the mechanism of drug composition,target and pathway was explored.Results:1.Overall:A total of 357 literatures were included,including 393 prescriptions,of which Taohong Siwu Decoction was the most widely used.The drugs were mainly used for promoting blood circulation and removing blood stasis,clearing heat,improving water and moistening moisture,tonifying and regulating qi.The four qi are mainly warm,the five flavors are mainly bitter,and the liver meridian is the majority.Angelica-Ligusticum chuanxiong is the high frequency drug pair and the core drug combination.2.Level:Among the syndrome of dampness and heat injection,Simiao Powder and Simiao Yong ’an Decoction were used most.High-frequency drugs were Radix Radix Cyranae Radix,Angelica Sinensis and Phaedocytes Phaedocytes,which mainly had cold and bitter properties.The most commonly used drugs for the syndrome of blood stasis and dampness obstruction were Angelica sinensis,Ligusticum chuanxiong and safflower,etc,with warm and sweet taste,and liver was the main drug channel.Seven new drug combinations were obtained.In the syndrome of spleen-kidney Yang deficiency,besides Astragalus membranaceus,Angelica sinensis and Ligusticum chuangusticum,aconite,cinnamon,Cornus,officinalis,Epimedium epimedium,Poria cocos and Atractylotractyloides appeared more frequently.Most of them were warm and sweet,and mainly belonged to the liver and spleen channels.Four new drug combinations evolved.Among the topical drugs,angelica sinensis,chuanxiong,safflower and other drugs to promote blood circulation and remove blood stasis were the main ones,mirabilite and borneol were used more,with mild and cold properties,bitter and spicy taste,and returned to the liver,heart and spleen meridians.12 new prescriptions were obtained.3.Analysis of the mechanism of action of high-frequency drug pairs:there were 64 active compound action targets of Angelica-Ligusticum chuanxiong,1653 disease targets,and 34drug-disease-related targets.The enrichment analysis of GO and KEGG pathways involved30 biological functions and 76 pathways.It is speculated that Angelica-Ligusticum chuanxiong may play a therapeutic role in deep vein thrombosis by affecting gene transcription,apoptosis process,oxidative stress response,hormone level,etc.Conclusions: The combination of TCMISS and data mining technology can summarize and analyze the rules of prescription drug use for diseases,predict the common target and pathway of drug and disease action,and provide reference for clinical treatment.
