Analysis Of Medication Rule Of Traditional Chinese Medicine For Behcet’s Disease Based On Complex Network And Data Mining Technology

Objective To explore the medication rules of traditional Chinese medicine in treating Behcet’s disease and provide reference for clinical treatment,based on the complex network and data mining method.Methods Prescriptions were extracted from literatures which were retrieved from CNKI,Wanfang,VIP,Sino Med and Pub Med database according to inclusion and exclusion criteria.Then,the database was established,and the data was standardized.Statistical analyses were performed using Microsoft Excle2016,Matlab2018 b,Gephi0.9.2 and IBM SPSS Statistics 25.0 for use frequency,efficacy,tropism of taste,primary medicines,association rules and Hierarchical cluster respectively.Results A total of 217 prescriptions for Behcet’s disease were screened out from 161 papers which including 225 Chinese herbs,involving 2314 times of use.From the perspective of efficacy classification,heat-clearing drugs(39.3%)are mostly used,followed by tonic drugs(27%),diuretic and dampening drugs(9.1%),blood promoting and stasis removing drugs(6.1%),and anti-epidermic drugs(4.7%),interior-warming drug(2.8%).The heat-clearing drugs included heat-clearing and damp-drying medicine(12.2%),heat-clearing and cooling blood medicine(11.6%),heat-clearing and detoxifying medicine(10.2%),heat-clearing and purging gunpowder(5.0%),and deficient heat clearing drug(0.3%);Deficiency drugs included Qi-tonifying drugs(14.6%),Blood-tonifying drugs(7.1%),Yin-tonifying drugs(4.4%),and Yang-tonifying drugs(0.1%).From the perspective of drug properties,the four qi of traditional Chinese medicine were mainly cold drugs,followed by heat,warm,calm,and cold;the five flavors were mainly sweet,bitter,and pungent;the main tropism of taste is the heart,liver,lung,spleen,stomach,and kidney.The complex network analysis revealed 49 primary Chinese medicines such as licorice,the Radix Rehmannia,Chinese angelica,red peony root,coptis,cork,tree peony bark,honeysuckle,scutellaria,poria cocos,and scrophulariaceae.The correlation analysis showed that the number of association rules between two,three,and four drugs are 10,15,and 6,respectively.The top 25 primary Chinese medicines were classified into three categories by cluster analysis.The first category included Bupleurum,Gardenia,Alisma,Radix Rehmanniae,Angelica,Ligusticum chuanxiong,and Licorice;the second category included red peony root,white peony root,scutellaria baicalensis,Coptis,Cork,Anemarrhena,Moutan Bark,Honeysuckle,Forsythia,Scrophularia,Salvia;the third category includes Poria,Atractylodes,Codonopsis,Smilax,Coix seed,Astragalus,Sophora flavescens.Conclusion Traditional Chinese medicine treatment of Behcet’s disease is mainly based on heat-clearing and detoxifying,heat-clearing and damp-drying,heat-clearing and cooling blood and Qi-tonifying drugs.Through the analysis of tropism of taste,it is concluded that the use of cold drugs in the treatment of Behcet’s disease in traditional Chinese medicine is the highest.The medicinal flavor is mainly sweet,bitter,and pungent.The meridian tropism is mainly heart,liver,lung,lung,spleen,stomach and kidney.Through association analysis and cluster analysis,the treatment of Behcet’s disease focuses on the basic pathogenesis of "wet","heat","toxic","stasis",and "deficiency".The treatment focuses on clearing away heat and detoxification,replenishing qi and dispelling dampness,cooling blood and promoting blood circulation.According to the different stages of the disease,the treatment has its own focus.In view of the above five major pathogenesis,three types of drug combinations are summarized: for the internal accumulation of damp-heat syndrome,Bupleurum,Gardenia,Alisma orientalis,Rehmannia glutinosa,Angelica,Ligusticum chuanxiong,licorice;for the heat intoying blood syndrome,red peony,white peony,Scutellaria baicalensis,Coptis,Cork,Anemarrhena,Moutan Bark,Honeysuckle,Forsythia,Scrophulariaceae,Salvia;for spleen deficiency syndrome type Poria,Atractylodes,Codonopsis,Smilax,Coix seed,Astragalus,Sophora flavescens.The use frequencies,complex networks,association rules,and systematic clustering in this study sorted out the prescriptions and medications of Behcet’s disease that have objective curative effects in the existing literature,which revealed some core drugs,strong association drug pairs and drug combination.It can provide some references and new ideas for clinical TCM treatment of Behcet’s disease,and it can also better guide the clinical practice of reasonable and effective prescription drugs.
