Objective:Observe the effect of Xiaojianzhong Decoction on the immune function of young rats with spleen deficiency and diarrhea,and preliminarily explore the mechanism of Xiaojianzhong Decoction in treating spleen deficiency and diarrhea.Methods: Forty male Kunming young mice were randomly divided into 4groups,10 in each group,which are A matched group,B splenic asthenia group,C Xiaojianzhong Decoction high-dose group(high-dose group),D Xiaojianzhong Decoction low-dose group(low-dose group).Except for the matched group Feed normal saline daily(0.2m1/10g),The other three groups were treated with Rhubarb decoction(0.2m1/10 g.d)for consecutive 7 days to establish spleen deficiency and diarontrol rhea model.When the model was built,the spleen deficiency group and the matched group were regularly fed normal saline(0.2m1/10 g.d),the high-dose group was fed Xiaojianzhong Decoction high-dose liquid(0.2m1/10 g.d),and the low-dose group was fed Xiaojianzhong Decoction low-dose liquid(0.2m1/10 g.d)for consecutive 10 days.Observe the general condition of the pups every day,weigh the pups of each group on the first day,the eighth day,and the 18 th day of the experiment.After the treatment,the orbital venous blood samples were collected.Serum immune indexes(C3,C4,IgA,IgM,IgG,IL-4,IFN-γ)were determined by enzyme-linked immunosorbent assay(ELISA);flow cytometry was used to detect each group of young rats The concentration of CD3 T,CD4T,and CD8 T in peripheral blood.Results:1.Changes in body weight of young rats Compared with the normal group,the weight of the pups in the spleen-deficiency group increased significantly(P<0.05).After the intervention of Xiaojianzhong decoction,Compared with the spleen deficiency group,the weight of the young mice in the high and low dose Xiaojianzhong Decoction groups increased,but the weight of the high dose Xiaojianzhong Decoction group increased significantly(P <0.01).2.Changes of serum C4 and C3 levels in young mice :Compared with macthed group,serum content of C4 pixu(spleen deficient)group was obviously increased(P < 0.01);Compared with group of pixu(spleen deficient),Xiaojianzhong soup in serum C4 content in high dose group were significantly lower(P < 0.01),and Xiaojianzhong soup serum C4 content in high dose group was obviously lower in low dose group(P <0.01).There was no significant change in serum C3 among all experimental groups(P> 0.05).3.Changes of serum IgA,IgM and IgG in young rats :Compared with the matched group,serum content of IgA,IgM and IgG pixu(spleen deficient)group were lower(P < 0.01,P < 0.05 and P < 0.01);Compared with group of pixu(spleen deficient),Xiaojianzhong medicinal broth in high and low dose group of serum IgA,IgM and IgG increased(P < 0.01);Serum IgA and IgG contents in Xiaojianzhong decoction high-dose group were higher than those in Xiaojianzhong decoction low-dose group(P<0.01,P < 0.05).4.Changes in serum IL-4 and IFN-γ concentrations of young mice :Compared with the matched group,the contents of serum IL-4 pixu(spleen deficient)IFN-gamma content decreased with increased significantly(P <0.01);Compared with group of pixu(spleen deficient),Xiaojianzhong medicinal broth in high and low dose group were lower serum IL-4content and IFN-gamma content increased(P < 0.01).5.Yong mice peripheral blood T lymphocyte subsets changes:Compared with matched group,the group of CD4 pixu(spleen deficient),CD3 T lymphocyte percentage,CD4 / CD8 ratios were significantly lower and CD8 T lymphocyte percentage was significantly increased(P < 0.01).Compared with group of pixu(spleen deficient),Xiaojianzhong medicinal broth CD4 T in the high and low dose group,CD3 T lymphocyte percentage,CD4 / CD8 ratio were significantly elevated and CD8 T lymphocyte percentage were significantly lower(P < 0.01);Xiaojianzhong medicinal broth in high dose group of CD4,CD3 T lymphocyte percentage,CD4 /CD8 ratios were higher than Xiaojianzhong medicinal broth in low dose group(P < 0.05,P < 0.01,P < 0.01),and CD8 T lymphocyte percentage significantly lower than the Xiaojianzhong medicinal broth in low dose group(P < 0.01).Conclusion:1.The spleen-deficiency and diarrhea infant mouse model was made by the rhubarb-bitter-cold diarrhea modelling method.After the modelling,the weight of the spleen-deficiency and diarrhea infant mice decreased significantly,and they showed loose stools,reduced food intake,inflexible exercise,dull fur,and curled up.,Indicating that the model was successful.Xiaojianzhong Decoction can increase the weight of spleen-deficiency and diarrhea young mice and improve the general condition of spleen-deficiency young mice.2.Diarrhea of pixu(spleen deficient)the content of serum IgA,IgG,IgM are reduced,and Xiaojianzhong Decoction can increase pixu(spleen deficient)diarrhea the content of serum IgA,IgG,IgM,diarrhea of pixu(spleen deficient)exist the humoral immune function decline,and Xiaojianzhong Decoction can improve the humoral immunity function of diarrhea of pixu(spleen deficient),and Xiaojianzhong Decoction in high dose group on the improvement of the humoral immune function is better than that of low dose group.3.The contents of serum IL-4 increased and serum IFN-γ decreased in the purgatory mice with spleen deficiency.However,Xiaojianzhong decoction can reduce the content of serum IL-4 and increase the content of serum IFN-γ to regulate the balance of Th1/Th2,suggesting that Xiaojianzhong decoction can promote the disorder of Th1/Th2 ratio to return to the normal level.4.The percentage of CD4,CD3 T lymphocytes and the ratio of CD4/CD8 were decreased while the percentage of CD8 T lymphocytes was increased.Xiaojianzhong decoction could increase the percentage of CD4,CD3 T lymphocytes and the ratio of CD4/CD8,and decrease the percentage of CD8 T lymphocytes in the young mice with spleen deficiency and diarrhea,but the effect of the high dose group was more than that of the low dose group.Tip cell immune function decline diarrhea were pixu(spleen deficient),tracey in soup can improve the cellular immune function of diarrhea were pixu(spleen deficient),and Xiaojianzhong decoction in high dose group in improving cellular immune function is better than that of low dose group.5.Diarrhea pixu(spleen deficient)there were specific immune function decline,Xiaojianzhong decoction can regulate diarrhea were nonspecific immune function of pixu(spleen deficient).This may be related to the treatment of spleen deficiency and diarrhea by Xiaojianzhong decoction and the improvement of spleen deficiency symptoms.The original dose of Xiaojianzhong Decoction Zhongjing(high dose)improves immune function better than the recommended dose(low dose)of modern textbooks.
