Objective:Exploring Professor Sun Wei’s medication rule in the treatment of chronic kidney disease,condensing the core prescription,observing the therapeutic effect,and exploring the gene target,protein and signal pathway between traditional Chinese medicine(TCM)and chronic kidney disease.Methods:Prospective and retrospective studies were conducted.Data mining methods was performed.First,we collected and standardized the data to establish database.Second,frequency statistics,association rules,complex networks and comparative method were exerted to interpret the experience of Professor Sun Wei of TCM on treatment of kidney diseases.Finally,we use network pharmacology to analyze the gene targets,proteins and signaling pathways of drug diseases.Results:Totally 225 cases of Professor Sun Wei in Jiangsu Province Hospital of TCM were involved.The frequency of herbs were analyzed.The common therapy was tonifying kidney,clearing heat and moisture,promotion of circulation.When Professor Sun treats different kidney diseases and different stages of disease,the treatment principle remains unchanged,but the focus of medication is different.In the treatment of CKD with the previous method,the total effective rate was 64%.Among them,patients with CKD stage 3-5 had more obvious effect.Combining"disease-symptoms-syndromes-laboratory tests-medicine",and integrated analysing to establish a novel efficacy evaluation system.After database screening and integration,we got 120 active ingredients,278 corresponding targets,440 gene targets of chronic kidney disease,and 70 targets of drug and disease.According to the visualized network diagram of "active ingredients-disease-targets",the most relevant active ingredients were quercetin,kaempferol and luteolin.The PPI network was analyzed,and the core sites such as ALB,AKT1,IL6,TNF,VEGFA and so on were screened.Go enrichment analysis showed that the biological processes such as gland development,endothelial cell proliferation,response to oxidative stress,reactive oxygen species metabolic process and cellular response to chemical stress had high correlation significance;The cell composition of membrane raft,membrane microdomain,membrane region,vessel lumen and secret granule lumen had strong correlation significance;Signaling receptor activator activity,receptor live activity,cytokine receptor binding,cytokine activity,phosphatase binding and other molecular functions have high correlation significance.The results of KEGG enrichment analysis showed that PI3K Akt signaling pathway,proteoglycans in cancer,human cytomegalovirus infection,AGE-RAGE signaling pathway in diabetic complications and prostate cancer had high correlation significance.Conclusion:1.The common TCM therapy of CKD is tonifying kidney,clearing and promoting blood circulation.Subsequently,the TCM pathogenesis of CKD is kidney deficiency,moisture and stasis.2.The different therapy strategy is dependent on the stage of kidney disease.3.Through computer algorithm and certification,inspection indicators and other associations,easy to better summarize the experience of the old Chinese medicine.4.It preliminarily verified the rationality and scientificity of the core prescription in the treatment of CKD.It is suggested that quercetin,kaempferol and luteolin are highly specific components,and PI3K Akt signaling pathway,AGE-RAGE signaling pathway and HIF-1 signaling pathway have high correlation significance.
