Objective:Based on the theory of "blood turbidity",according to the basic pathogenesis characteristics of turbid pathogen and unclear blood of hyperlipidemia,to observe the clinical efficacy of Huazhuo Xingxue decoction in the treatment of hyperlipidemia,and to further explore the effect of Huazhuo Xingxue decoction on DGAT2 m RNA expression and lipid metabolism in hyperlipidemia model rats,so as to provide scientific basis for the treatment of hyperlipidemia with Huazhuo Xingxue decoction.Methods:1.Sixty patients with hyperlipidemia who met the inclusion criteria were randomly divided into treatment group(n = 30)and control group(n = 30).The treatment group was treated with routine treatment and Huazhuo Xingxue decoction,while the control group was treated with routine treatment and Xuezhitong capsule.Both groups were treated for 4 weeks.The changes of laboratory indexes and TCM syndrome quantitative scores of the two groups before and after treatment were observed,and the respective curative effects of the two groups were evaluated.2.Through experimental study,using the method of real-time fluorescence quantitative polymerase chain reaction(Real-time PCR)to observe the effect of Huazhuo Xingxue decoction on lipid metabolism and DGAT2 m RNA expression in liver tissue of hyperlipidemia model rats,and to further explore the mechanism of intervention of Huazhuo Xingxue decoction on blood lipid level.Results:In the aspect of clinical trial,both the treatment group and the control group could reduce the levels of TC,TG and LDL-C in patients with hyperlipidemia,and the effect of the treatment group was better than that of the control group.At the same time,the treatment group also increased the level of HDL-C,and the difference was statistically significant(P < 0.01).In terms of laboratory index evaluation,the curative effect of the treatment group was better than that of the control group,and the difference was statistically significant(P<0.05).In the evaluation of TCM syndrome,the curative effect of the treatment group was better than that of the control group,and the difference was statistically significant(P<0.01).In terms of clinical trial research,both the treatment group and the control group can reduce the TC level of patients with hyperlipidemia,and the effect of the treatment group is better than that of the control group.At the same time,the treatment group can also reduce the level of TG and LDL-C in patients and increase the level of HDL-C in patients.The effect of level,the difference is statistically significant(P<0.01,P<0.05).Compared with high and low dose groups,TC and TG decreased and HDL-C increased more significantly in the middle dose group of Huazhuo Xingxue decoction(P<0.05).The expression of DGAT2 m RNA in the liver tissue of the model group was higher than that of the blank group,and compared with the model group,the expression of DGAT2 m RNA in the control group,positive drug group and high,middle and low dose groups of Huazhuo Xingxue decoction decreased(P<0.05,P<0.05),and the expression of DGAT2 m RNA in the middle dose group was lower than that in the high and low dose groups(P<0.05).Conclusion:Huazhuo Xingxue decoction can effectively improve the blood lipid level of patients with hyperlipidemia,and the prescription has no obvious side effects,and its mechanism may be related to down-regulating the expression of DGAT2 m RNA in liver tissue.It is proved that Huazhuo Xingxue decoction is an effective prescription for clinical treatment of hyperlipidemia,which is worthy of further promotion.
