| Objective: To observe the effects of Yangganyishui Granules on blood pressure,urinary ACR,β2-MG,serum SCR,BUN,AngⅡ levels and kidney pathology in SD rats induced by high-salt and high-fat diet;and kidney tissue AngⅡ,TRPC6 and NF-κB Analyze the effects of protein expression levels in the early stage of hypertension to reduce blood pressure and protect the target of renal damage,in order to develop Yanggan Yishui Granules as a prescription for protecting early renal damage of hypertension.Methods: 50 SD rats were randomly divided into blank group,model group,Yangganyishui Granules group(YGYS group),Benazepril group(ACEI group),Yangganyishui Granules + Benazepril group(YGYS+ACEI group),10 in each group.The YGYS group,ACEI group,and YGYS+ACEI group were given more salt and fat food for 6 weeks.After 6 weeks,the YGYS group was given Yangganyishui Granule Suspension(2.7g crude drug/kg/d),and the ACEI group was given Benazepril suspension(0.9mg/kg/d),gavage 1 time/d;YGYS+ACEI group were given Yangganyishui Granule Suspension(2.7g crude drug/kg/d)and Benazepril suspension(0.9mg/kg/d),gavage 2 times/d;two other groups feed the same drench water once per day;the period was 4 weeks.Test blood pressure for rats every week;collect the urine ACR and β2-MG of each group before and after the administration for quantitative detection;take blood samples from the rats through the abdominal aorta to detect the contents of SCR,BUN and AngⅡ in the serum;Retain specimens of rat kidneys for detection of AngⅡ,TRPC6,NF-κB and other proteins in kidney tissues;watch the sick changes of rat renal tissue.Results: 1 Blood pressure: Ratio to the blank group,YGYS group,ACEI group,ACEI+YGYS group modeled for 6 weeks,the SBP increased significantly(P<0.01),though the SBP among groups have no distinction(P>0.05).After remedy,the SBP in the YGYS group,ACEI group,and ACEI+YGYS group were lower(P<0.01).Ratio to the ACEI group,the SBP of YGYS group was rised compared to ACEI group(P<0.05),and the SBP of ACEI+YGYS group was reduced compared to ACEI group(P<0.05).2 Urinary ACR and β2-MG: Ratio to the blank group,the untreat group,YGYS group,ACEI group,ACEI+YGYS group SD rats modeled for 6 weeks,urine ACR,β2-MG increased(P <0.01).After 4 weeks of administration,compared with the model group,the urinary ACR and β2-MG of SD rats in the YGYS group,ACEI group,and ACEI+YGYS group decreased(P<0.01).Compared with the ACEI group,the urinary ACR and β2-MG of SD rats in the YGYS group were higher than those in the ACEI group(P < 0.05),and the urinary ACR and β2-MG of SD rats in the ACEI+YGYS group were lower than those in the ACEI group(P<0.05).3 Serum SCR and BUN: Ratio to the blank group,serum SCR and BUN in the untreat group,YGYS group,ACEI group,YGYS+ACEI group have no diversity(P>0.05).4 Serum AngⅡ: Ratio the blank group,the serum AngⅡ of the untreat group,YGYS group,ACEI group,YGYS+ACEI group increased(P<0.05).Ratio to the model group,the serum AngⅡ of YGYS group,ACEI group,and YGYS+ACEI group all decreased(P<0.05).Compared with the ACEI group,the serum AngⅡ of SD rats in the YGYS group was rised contrast to the ACEI group,and the serum AngⅡ of ACEI+YGYS group was reduced tcontrast to the ACEI group,but there was no significant change(P>0.05).5 Pathway protein expression level: Compared with the blank group,the expression levels of AngⅡ,TRPC6 and NF-κB protein in the kidney tissue of the model group,YGYS group,ACEI group,YGYS+ACEI group go up evidently(P<0.01).Ratio to the model group,the standard of AngⅡ,TRPC6 and NF-κB in the kidney of the YGYS group,ACEI group,and YGYS+ACEI group reduced(P<0.01).Ratio to the ACEI group,t AngⅡ,TRPC6 and NF-κB in the kidney tissue of the YGYS group were over the ACEI group,but there was no significant difference(P>0.05).The AngⅡ,TRPC6 and NF-κB in the kidney tissue of the ACEI+YGYS group The protein expression level was lower than that of the ACEI group,and there was a difference(P<0.05).6 HE staining observation of kidney tissue: The structure of kidney tissue of SD rats in the blank group was normal.In the model group,some glomeruli were seen atrophy;nearby glomeruli were compensated hypertrophy,the number of mesangial cells increased significantly,the glomerular cyst cavity was abnormal,and the basement membrane was thickened;some renal tubular epithelial cells were edema and cytoplasm loose.After treatment,rats in the YGYS group,ACEI group,and YGYS+ACEI group all improved the above-mentioned performance to varying degrees.Conclusion: Yangganyishui Granules can alleviate the early renal damage of high-salt and high-fat diet-induced hypertension in SD rats.The mechanism may be that it inhibits the AngⅡ/TRPC6/NF-κB pathway. |
