| Objective: Preliminarily explore whether the cultivated Paeonia lactiflora Pall.can be processed into Paeoniae Radix Rubra(PRR)for use.This article combs through the relevant content in the Chinese herbal books,all editions of the pharmacopoeia and recent literatures,hoping to find the supporting basis for the processing of cultivated paeonia lactiflora into PRR.Taking the Bozhou cultivated paeonia lactiflora as the research object,the cork,phloem and xylem of the roots of Bozhou cultivated P.lactiflora Pall.are studied to explore the possible influence of "peeling" processing on the chemical composition of the medicinal materials.PRR and Paeoniae Radix Alba(PRA)processed by Bozhou cultivated P.lactiflora Pall.(PRR-B,PRA-B)are compared with Neimenggu wild P.lactiflora Pall.(PRR-N)in terms of chemical composition and blood-activating effect,explore the difference of chemical composition and the effect of promoting blood circulation and discuss whether the Bozhou cultivated P.lactiflora Pall.can be processed into PRR.Explore the rational connotation of processing cultivated P.lactiflora Pall.into PRR.Methods: Sort out the records of “shaoyao”,“baishao” and “chishao” in Chinese herbal books,all editions of the pharmacopoeia and recent literatures,clarify the origin,evolution and efficacy of the separate use of PRA and PRR.Randomly take fresh roots of Bozhou P.lactiflora Pall.,peel off three parts of cork,phloem and xylem,dry,crush,and ultrasonically extract with 50% ethanol.The chemical composition of the three parts was compared using the method of HPLC characteristic chromatogram,and the chemical composition of the three parts was further qualitatively and quantitatively quantified by UPLC-MS.The water extracts of PRR-B,PRA-B and PRR-N were tested for multi-component content by HPLC method.The water extract was given to the experimental blood stasis model rats by gavage,and the differences in the effects of promoting blood circulation were compared by measuring the indexes of hemorheology and coagulation function of the rats.Result: 1.The processing of cultivated paeonia lactiflora into PRR is in compliance with the 2020 Chinese Pharmacopoeia.According to the record of the red flower’s one can be processed into chishaoyao in some Chinese herbal books,Bozhou cultivated paeonia lactiflora with red flowers can also be processed into PRR.But they do not conform to the view in modern herbal books that PRR is mostly wild products from the northern and PRA is mostly cultivated products in the south.2.The characteristic chromatogram of the cork,phloem and xylem from the Bozhou cultivated paeonia lactiflora root have been established.The chromatograms’ similarity between samples of any part(within the group)is not less than 0.900,indicating that the chemical composition of the same part is relatively similar,among which the components in the xylem group are the closest,the phloem group is the second,and the cortex group is less consistent.The similarities between cork and phloem,cork and wood,and phloem and wood(between groups)are respectively0.841 ~ 0.974,0.586 ~ 0.766,and 0.653 ~ 0.928,it can be seen that the chemical composition of different parts is different,and the difference between cortex and xylem is the largest.3.525 differential compounds were screened between cork and phloem,452 differential compounds between cork and xylem,and 328 differential compounds between phloem and xylem.The abundance analysis of the differential compounds between the three comparison groups,and the top 50 compounds with large differences were obtained.The correlation coefficients of the first 50 different compounds were obtained by correlation analysis.The top 20 metabolic pathways were listed by the enrichment analysis of metabolic pathways for different compounds with larger enrichment factors among the three comparison groups.4.An HPLC method was established for the simultaneous determination of gallic acid,oxypaeoniflorin,catechin,albiflorin,paeoniflorin,1,2,3,4,6-pentagalloyl glucose and benzoic acid.The contents of 7 components in the water extracts of PRA-B,PRR-B and PRR-N were determined.Gallic acid: PRR-N > PRR-B > PRA-B;oxypaeoniflorin,paeoniflorin and 1,2,3,4,6-pentagalloyl glucose: PRR-N > PRA-B >PRR-B;catechins in PRR-B and PRA-B did not reach the detection limit,but PRR-N has the highest catechin content;albiflorin: PRA-B > PRR-B > PRR-N;benzoic acid:PRR-B > PRR-N > PRA-B.5.Under the same crude drug dosage,the three medicinal water extracts of PRA-B,PRR-B and PRR-N have different degrees of improvement on the blood rheology and coagulation function of the liver depression and cold coagulation blood stasis in SD rats caused by this experiment.Reduce red blood cell aggregation:PRA-B > PRR-B > PRR-N;there is no difference in the effect of reducing plasma viscosity;weaken erythrocyte sedimentation rate: PRA-B = PRR-B > PRR-N;extend PT: PRA-B = PRR-B > PRR-N;all three can prolong APTT,reduce FBG content,and prolong TT,and there is no difference in the effects of them.Conclusion: Bozhou cultivated P.lactiflora Pall.can be processed into PRR under the premise of complying with the provisions of the pharmacopoeia.Except for the glycerolphosphate,the abundance of the other chemical components in the cortex of Bozhou cultivated P.lactiflora Pall.root is mostly lower than that of the phloem.So the treatment of scraping off the cork may have little effect on the chemical composition of the medicinal materials.The contents of the 7 components between PRR-B and PRA-B are relatively close,and are quite different from those of PRR-N.For the blood stasis model in this article,the effect of PRR-B is slightly weaker than that of PRA-B,but slightly stronger than that of Inner PRR-N.Therefore,it remains to be discussed to processing the root of Bozhou cultivated P.lactiflora Pall.into PRR. |
