Objective:1 The quality evaluation system of Paeoniae Radix Alba,slices and standard decoction were established,and to exploring the processing method of Paeonia Radix Alba for the quality transmission of Paeoniae Radix Alba,slices and standard decoction.The specific parameters of each processing link are optimized to provide a basis for preparing Paeoniae Radix Alba,slices and standard decoction.2 Study on the mass transfer regularity of Paeoniae Radix Alba,slices and standard decoction.Methods:1 Using theappearance traits and paeoniflorin content as indicators,explore the influence of factors such as the origin of white peony roots and the softening of white peony roots on the quality of white peony roots and slices.Simultaneously determine the content of four monoterpene glycoside components in standard decoction by HPLC method,and calculate the transfer rate in standard decoction of Paeoniae Radix Alba with different thickness,combined with blanking rate,warping rate,chipping rate and extract yield,to explore the effect of slice thickness on the quality of standard decoction.2 The 20 batches of Paeoniae Radix Alba from Bozhou were processed into 20 batches of Paeoniae Radix Alba,20 batches of w Paeoniae Radix Alba slices and 10 batches of standard decoction.TLC analysis of Paeoniae Radix Alba and Paeoniae Radix Alba slices,content determination and fingerprint analysis of Paeoniae Radix Alba,Paeoniae Radix Alba slices and standard decoctions.Results:1 The optimized TLC conditions of Paeoniae Radix Alba have good specificity and durability;four content determination methods and HPLC fingerprint method have been established,and their methodology is well investigated.2 The processing of Paeoniae Radix Alba,softening of Paeoniae Radix Alba and slice thickness have obvious effects on the quality of Paeoniae Radix Alba.The preferred process of Paeoniae Radix Alba is to take fresh Paeoniae Radix Alba of equal size,wash it,put in twice the amount of 90 ℃ water and cook for 10 minutes,take it out into cold water,cool it,peel it in a drum,and dry it at 60 ℃ in a low temperature dryer.The best process for the processing of sliced pieces is to take Paeoniae Radix Alba of equal size,choose to soak them for 24 hours,moisten them for 40 minutes,cut them into thin slices,and dry them in an oven at 60 ℃ for 11 hours.The blanking rate,warping rate,and chipping rate of Paeoniae Radix Alba decreased with the increase of the thickness;the extract yield and transfer rate of each component of Baishao decoction were inversely proportional to the thickness of standard decoction.3 In the TLC of Paeoniae Radix Alba,the spots of paeoniflorin are purplish red.There is no obvious difference between the TLC characteristics of Paeoniae Radix Alba and slices.The average content of paeoniflorin is 2.979%,the content of albiflorin is 0.635%,the content of oxypaeoniflorin is 0.025% and the content of benzoylpaeoniflorin is0.084% in 20 batches of Paeoniae Radix Alba.The average content of paeoniflorin is2.573%,the average content of albiflorin is 0.480%,The average content of oxypaeoniflorin is 0.022% and the content of benzoylpaeoniflorin is 0.074% in 20 batches of Paeoniae Radix Alba slices.The content of paeoniflorin was 2.244%,the content of albiflorin was 0.374%,The average content of oxypaeoniflorin is 0.018% and the content of benzoylpaeoniflorin is 0.062% in 10 batches of standard decoction.During the process from Paeoniae Radix Alba to pieces,the average transfer rate of paeoniflorin was 85.13%,the average transfer rate of paeoniflorin was 78.65%,the average transfer rate of oxypaeoniflorin was 88.74%,and the benzoylpaeoniflorin The average transfer rate is 87.64%;in the process of Paeoniae Radix Alba slices to standard decoction,the transfer rate of paeoniflorin is 86.66%,the transfer rate of paeoniflorin is78.42%,and the transfer rate of oxidized paeoniflorin is average The value is 84.44%,the average transfer rate of benzoyl paeoniflorin is 73.71%;the transfer rate of paeoniflorin is 72.36% in the whole process of Baishao medicine to Baishao decoction standard soup,and the transfer rate of paeoniflorin 59.96%,the average transfer rate of oxidized paeoniflorin is 76.34%,and the average transfer rate of benzoyl paeoniflorin is63.33%.There are 22 common peaks in the fingerprint of Paeoniae Radix Alba,14 common peaks in the fingerprint of Paeoniae Radix Alba slices,and 10 common peaks in the fingerprint of standard decoction.There are 10 peaks in all.The peaks are 1,4,6,8,9,10,13,15,18 and 20.Compared with the chromatogram of the control sample,peaks 6,9,10 and 20 are oxypaeoniflorin,albiflorin,paeoniflorin and benzoylpaeoniflorin,respectively.Comparing of the Paeoniae Radix Alba and its pieces,the number of peaks of Paeoniae Radix Alba is 3,5,11,12,14,16,17 and 19.Comparing of the Paeoniae Radix Alba and standard decoction,Paeoniae Radix Alba have peaks of 2,3,5,7,11,12,16,17,19 and 21.Comparing of Paeoniae Radix Alba pieces and standard decoction,Paeoniae Radix Alba pieces have peaks of 2,7,21 and 22.Conclusion:1 The established quality evaluation system of Paeoniae Radix Alba-slices-standard decoction is stable and reliable,and can comprehensively evaluate the quality changes during preparation of Paeoniae Radix Alba-slices-standard decoction.2 TLC of Paeoniae Radix Alba and its pieces preferred in this experiment is stable and feasible.It is the production of white peony root and decoction piece.3 The quality variation of the preparation process of Paeoniae Radix Alba-slices-standard decoction was studied,which provided a reference for the quality control during the processing of Paeoniae Radix Alba and slices.
