Colorectal cancer(CRC)is the fifth most common type of cancer in China.Its pathogenesis has not been fully understood yet.Honokiol(HNK)is extracted from the bark and branches of magnolia family Magnolia officinalis.It has many proven pharmacological effects,including anticancer,antibacterial,antiviral activities,protecting the nervous system.The present study aimed to investigate the molecular mechanisms and inhibitory effect of HNK on the proliferation and apoptosis of colon cancer SW620 cells.Crystal violet staining,flow cytometry and western blotting techniques were used for the detection of the inhibitory effect of HNK on the proliferation and apoptosis of SW620 cells and its possible molecular mechanisms.The results revealed that HNK significantly inhibited the proliferation of SW620 cells and induced apoptosis,and up-regulated the expression levels of TGF-β1.The western blot analysis showed that the HNK up-regulated the expression levels of phosphorylated p38 in SW620 cells.Meanwhile,the HNK up-regulated the protein levels of phosphorylated YAP.The overexpression of exogenous adenoviruses TGF-β1 and TGF-β1 inhibitors significantly enhanced and inhibited this effect,respectively.In conclusion,HNK has obvious anti-cancer activities,which might be due the up-regulating expression of TGF-β1 and phosphorylated YAP by activating the p38 MAPK signaling pathway.
