In many anti-tumor Chinese medicine effective components,Honokiol(HNK)shows strong anti-tumor activity.However,it’s detailed mechanism is still unclear.In our research,we have validated Honokiol could inhibit growth and induce apoptosis of HCT116 cells in a time and concentration dependent manner by using some basic molecular biology technologies.Further experiments shows that Honokiol could up-regulate the protein level and mRNA level of BMP7 in HCT116 cells,which similar to the results of Honokiol on SW620 and Lo Vo cells.Recombinant adenovirus technology was used to make AdBMP7,which could strengthen Honokiol’s effect further;While its anti-body partly attenuated Honokiol’s effect.Besides,the expression of p53 is positively associated with the concentration and actuation duration of Honokiol.And the results are also similar to the influence of Honokiol on SW620 and LoVo cells.Similarly,p53 adenovirus could strengthen Honokiol’s effect,while its inhibitor partly reversed Honokiol’s effect and induce cells’ growth.Mechanism analysis: Honokiol has no substantial effect on the expression of Smad1/5/8 and phosphorylated Smad1/5/8 in HCT116 cells,which indicates Honokiol exert its effect not through canonical BMPs/Smad signal pathway.The results showed BMP7 may mediates the anticancer effect of Honokiol by up-regulating p53 in colon cancer cells.
