| Colon cancer is one of the most common malignant tumors of the digestive tract.Its pathogenesis is still unclear,which has caused great obstacles to its treatment and prognosis.Therefore,it’s urgent to develop effective anticancer drugs.Traditional Chinese medicine or herbal derivatives are currently widely used as anti-tumor adjuvant drugs.Honokiol is a natural compound extracted from the bark and leaves of Magnolia officinalis.It is widely studied for its multiple pharmacological activities,such as anti-cancer,anti-oxidation,anti-angiogenesis,anti-inflammatory,but its anticancer mechanism remains unclear.In this study,we used biological techniques to investigate the anticancer activity of honokiol in colon cancer cells and to analyze its anticancer mechanism.Our study found that honokiol has a good inhibitory effect on several colon cancer cell lines,and SW620 cells are more sensitive to it.Honokiol can inhibit cell proliferation,arrests cell cycle arrest at G1 phase,and induce apoptosis.In the mechanism part,it was found that honokiol activated BMPs signaling and up-regulated the expression of BMP7.Overexpression of BMP7 enhanced the anti-cancer effect of honokiol,silencing BMP7 significantly attenuated the effect of honokiol.Honokiol has no significant effect on the classical BMPs signaling pathway,but reduces the activity of the PI3K/ Akt signal in SW620 cell lines.The activity of BMP7 can affect the regulation of honokiol on PI3K/ Akt.In addition,honokiol reduced the phosphorylation of PTEN,which was also affected by the activity of BMP7.Our research confirms that honokiol may be a potential anti-colon cancer drug.BMP7 mediates the anticancer activity of HNK by enhancing the function of PTEN,which partially reduces the activity of the PI3K/ Akt signaling pathway.It provides a new direction and theoretical basis for the development of therapeutic drugs for colon cancer. |
