| Objective To explore the expression of TFF3 in cervical cancer C33 A cells,at the same time to investigate the effect of TFF3 overexpression on proliferation and migration of cervical cancer cells.Methods In vitro cultivation of cervical cancer C33 A cells,using liposome transfection method,the overexpression vector and its negative control of TFF3 were transfected into cervical cancer C33 A cells.The experiment was divided into three groups,respectively in TFF3 overexpression group(C33A-OE-TFF3 group)and the negative control group(C33A-OE-NC group),blank control group(C33A-control group).QPCR method detected the expression level of TFF3 gene in C33A-OE-TFF3 group and C33A-OE-NC group and C33A-control group.CCK-8 method was used to detect the effect of TFF3 gene on proliferation ability of cervical cancer cells.Scratch test was used to detect the effect of TFF3 gene on migration ability of cervical cancer cells.Results1.QPCR method showed that the expression level of TFF3 gene of C33A-OE-TFF3 group was significantly higher than the negative control group and blank control group.The difference was statistically significant(P<0.001).2.The result of CCK-8 showed that observed in 24 h and 48 h and 72 h,the optical density of C33A-OE-TFF3 group was higher than C33A-OE-NC group and C33A-Control group.C33 A cells’ proliferation ability of C33A-OE-TFF3 group enhanced significantly in72 h.The difference was statistically significant(P<0.001).And compared with blank control group and the negative control group,the difference was not statistically significant(P>0.05).3.The result of scratch test showed that,observed in 24 h,migration ability of C33A-OE-TFF3 group was higher than C33A-OE-NC group and C33A-Control group.The difference was statistically significant(P<0.05).Compared to blank control group and the negative control group in 48 h,C33A cells’ migration ability of C33A-OE-TFF3 group enhanced obviously.The difference was statistically significant(P<0.01).And compared with blank control group and the negative control group,the difference was not statistically significant(P> 0.05).Conclusion1.To build successfully TFF3 overexpression vector,cervical cancer C33 A cells of TFF3 overexpression proliferation and migration ability enhance.2.TFF3 can promote the occurrence and development of cervical cancer. |
