| Objective:To study the effects of Linggui Zhugan Added Decoction combined with captopril on serum BNP,Endothelin-1(ET-1)and Caspase-3(Caspase-3)levels of chronic heart failure rats,which provides a scientific basis for clinical medication.Methods:1.Selecting 50 SPF male SD male rats and dividing them into normal group(10)and control group(40)for modeling.In the healthy group,normal saline was injected into the abdominal cavity,and the heart failure group is continuously intraperitoneally injected with isoproterenol 5mg/(kg·d).A rat model of chronic heart failure with LVEF≤50%was established within 7 days,and the model was successfully established.2.Using random number table method,40 SPF SD rats with heart failure were divided into heart failure group,Linggui Zhugan Added group(Chinese medicine group),captopril(western medicine group),Linggui Zhugan Added group combined with captopril group(Chinese and Western medicine group).According to the calculation of the "Pharmacological Experimental Methodology" to the rats to give the drug dose,the rats can be given 6.3 times the human dose at the same time every day.Linggui Zhugan Added group was intragastrical administered at a crude drug amount of 3g/kg/d and captopril 2.5mg/kg/d.The healthy group and the heart failure group received the same amount of normal saline for 30 days.3.After 30 days of continuous intragastric administration in all groups,observing the morphology and movement of the rats,performing cardiac echocardiography and retaining the data,weighing the rats,drawing blood from the abdominal aorta,taking out the heart,weighing and calculating the heart mass index.After the heart tissue was cut,HE staining was performed,and the changes of the myocardial pathological images of each group were observed under an optical microscope.4.The serum levels of BNP,ET-1,and Caspase-3 in rats were determined by ELISA,and the heart mass index was calculated.The SPSS25.0 software was used for overall statistical evaluation and data analysis.Result:1.General state of rats:Compared with the heart failure group,the three gavage treatment groups can improve the performance of heart failure rats such as fatigue,dyspnea,anorexia,hair loss,edema,etc.The rats in the combination of Chinese and Western medicine are in the best state,there was no significant difference between the Linggui Zhugan Added group and the captopril group.2.Cardiac color ultrasound:According to the data analysis of LVEF and heart weight index,it can be concluded that compared with the heart failure group,the three gavage treatment groups can help to improve the heart function of rats(P<0.05),and the combination of Chinese and Western medicine group The curative effect was the best(P<0.05),but there was no significant difference among the three gastric administration groups(P>0.05).3.BNP level:Compared with the heart failure group,the BNP level of the three groups of intragastric administration group was significantly reduced(P<0.05).The difference between the groups was obtained from the comparative analysis.The treatment effect of integrated traditional Chinese and western medicine was the best(P<0.05).4.ET-1 and Caspase-3 levels:Compared with the heart failure group,the ET-1 and Caspase-3 levels of the three gavage treatment groups were significantly lower(P<0.05).The difference between the two groups can be compared with the combination of Chinese and Western medicine.This group has the best curative effect(P<0.05).5.The examination of myocardial HE staining microscope confirmed that compared with the normal group,the cardiomyocytes of the other four groups of heart failure rats had cardiomyocyte hypertrophy,thickened muscle cell fiber structure,enlarged nucleus and inner nuclear space,and disordered arrangement.Cardiomyocytes may undergo degeneration such as valvular necrosis and vacuole,accompanied by inflammatory cell infiltration of different severity.After medication,the degree of myocardial cell damage is alleviated.Conclusions:1.Linggui Zhugan Added Decoction combined with Captopril can effectively improve the general state of rats with heart failure and reduce the serum levels of BNP,ET-1 and Caspase-3 in rats with chronic heart failure.The therapeutic effect of Lingguizhu Ganzengwei Decoction is similar to that of captopril,but two Combination of drugs has the best effect.2.Linggui Zhugan Added Decoction combined with Captopril can increase the cardiac ejection fraction of rats with heart failure,reduce the heart mass index,and improve the heart function of rats with heart failure.3.Linggui Zhugan Added Decoction combined with Captopril can alleviate myocardial cell damage in rats with chronic heart failure and effectively treat rats with chronic heart failure. |
