| The main chemical constituents of Melilot are proteins,amino acids,oils,coumarin,saponins,polyphenols,polysaccharides,flavonoids and alkaloids and other biologically active components,which are widely used in agricultural medicine application.In this paper,the melilotus officinalis was selected in Quanzi Street,Jimsar County,Xinjiang,China,and the total flavonoids,total amino acids,total coumarin and total polyphenols in the whole plant,stem,leaf and flower of the melilotus officinalis were determined.The whole plant,stem,leaf and flower were analyzed by chromatographic analysis of 3 kinds of anions,and the trace elements in the whole plant,stem,leaf and flower were analyzed by flame atomic absorption spectrometry.The main contents of this paper are as follows:1.Microscope was used to observe the powder tissue and sliced tissue of melilotus officinalis.Observation results showed that there were calcium oxalate cubic crystals arranged on the fiber of melilotus officinalis powder tissue;the stem cortex of the sliced tissue was symmetrically distributed in the epidermis,and the leaf veins and vascular bundles were distributed axisymmetrically.2.The mid-infrared spectral characteristics of the whole plant,stems,leaves and flowers of melilotus officinalis were analyzed,and the experimental results were analyzed;Through the comparative analysis of ultraviolet spectral lines of the extract of ymelilotus officinalis extract of three different solvents of petroleum ether,methanol and water,it was shown that the ultraviolet spectroscopy of melilotus officinalis can be used as one of the qualitative identification methods.3.The total flavonoids of the whole grass,stem,leaf and flower of the whole grass,stem,leaf and flower of the melilotus officinalis were determined by colorimetry of aluminum chloride-sodium acetate buffer,and the test results showed that the total flavonoid content of the rhinoceros leaf of the yellow flower tree was as high as 22.05mg/g,the lowest total flavonoid content in the stems of melilotus officinalis was 1.32mg/g;the total polyphenols content of the whole plant,stems,leaves and flowers of four different parts of melilotus officinalis were determined by the Folin phenol colorimetric method.The results showed that the total polyphenol content of the melilotus officinalis flower was 16.76 mg/g at the highest,and the total polyphenol content of the melilotus officinalis stem was the lowest at 3.60 mg/g.The content of total amino acids in four different parts of stems,leaves and flowers was determined,and the results showed that the total amino acid content of the melilotus officinalis was 1.03 mg/g,the lowest was 0.20 mg/g;The total coumarin content of the whole plant,stem,leaf,and flower of the flower and grass was determined.The results showed that the total coumarin content of the leaves of the melilotus officinalis was the highest at 32.26 mg/g.The minimum element content is 10.87 mg/g.The results showed that there were some differences in the total component contents of the whole plant,stem,leaf and flower of Melilot,and the total coumarin content of melilotus officinalis was the highest.4.Using high performance liquid chromatography,methanol(A):water(B)was selected as the mobile phase,and the content of coumarins in four different parts of the whole plant,stem,leaf,and flower of the melilotus officinalis was determined.The content of coumarin in the flower of Melilot is the highest at 0.564 mg/g,the content in leaves is second only to the content of coumarin in the flower,at 0.458mg/g,and the content of coumarin in the whole plant is the lowest at 0.176 mg/g.The experimental results provide basic theoretical data for the future scientific research of melilotus officinalis.5.Ion chromatography and flame atomic absorption spectrometry were used to determine the anions and trace elements of the whole plant,stem,leaf,and flower of the melilotus officinalis.Ion chromatography was used to determine Cl-,NO3-,SO42-in four different parts of the whole plant,stem,leaf and flower of melilotus officinalis.The measured results showed that the content of three anions in the leaves of melilotus officinalis was the highest,which was 5.496 mg/g,0.197 mg/g,5.413mg/g,the Cl-and NO3-contents of the whole plant of the melilotus officinalis were the lowest,which were 1.604 mg/g and 0.026 mg/g,respectively,and the SO42-content of the melilotus officinalis stem was the lowest,which was 1.412 mg/g;the content of Fe,Mn and Cu in four different parts of the whole grass,stems,leaves and flowers of Melilot was determined by flame atomic absorption spectrometry.were2.043 mg/g,2.035 mg/g,and 2.589 mg/g,and the contents of three trace elements were the lowest in the stems of the melilotus officinalis,which were 0.335 mg/g,0.354 mg/g,and 0.277 mg/g in sequence.The results of this experiment provide a scientific basis for the research on medicinal anions and trace elements of the melilotus officinalis. |
