| Oudemansiella raphanipes is a rare artificial cultivar with high nutritional and medicinal value.However,the residue is often discarded,which not only wastes a lot of available substances,but also easily causes secondary environmental pollution.In this study,O.Raphanipies residue polysaccharide(ORP)was obtained by water extraction and alcohol precipitation.Its structure was preliminarily analyzed,its antioxidant activity was detected in vitro,and the effect of ORP on hepatoenteric disease of lipopolysaccharide(LPS)-induced mice was verified.The results were as follows:(1)ORP structure analysis.The molecular weight of ORP determined by Gel permeation chromatography(GPC)was 1.34×10~4Da.The results of high performance liquid chromatography(HPLC)showed that ORP contained mannose,ribose,rhamnose,glucuronic acid,galacturonic acid,glucose,galactose,xylose,arabinose,fucose,and the mass fractions were 14.23%,4.98%,4.16%,5.26%,0.37%,18.91%,23.04%,10.87%,13.25%,4.92%,respectively.Fourier transform infrared spectroscopy(FT-IR)analysis showed that ORP was pyranose withβglycosidic bond.(2)Antioxidant activity of ORP in vitro.ORP at 800mg/L concentration had good scavenging ability on DPPH,and the scavenging rate reached 77.37±2.83%;the scavenging rate on superoxide anion reached 77.46±2.74%at 1000mg/L concentration,and the scavenging ability of hydroxyl radical reached 73.36±2.59%and reducing power reached1.74±0.07.The results showed that ORP had significant antioxidant activity in vitro.(3)Protective effect of ORP on liver and intestine.(1)ORP can alleviate the increase of liver and intestine weight caused by inflammatory edema,and reduce the phenomenon of liver and intestine damage;(2)ORP can decrease the activities of alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP)in serum.It protects the liver and effectively relieves inflammation.(3)ORP decreased the levels of IL-6,IL-10,IL-1βand TNF-αin liver and intestinal tract,and protected the liver intestinal tract from LPS damage.Meanwhile,superoxide dismutase(SOD)and catalase(CAT)in liver intestinal tract were detected.The activities of CAT and glutathione peroxidase(GSH-Px)were significantly increased,while malondialdehyde(MDA)was inhibited.The results showed that ORP can alleviate inflammatory effects by regulating the activity of antioxidant enzymes and the content of inflammatory factors,thus having a protective effect on the liver and intestine.In addition,morphological observation of liver and intestinal tissues showed that ORP could significantly reduce l PS-induced hepatointestinal injury.Above all,ORP in vitro antioxidant capacity significantly,and can be through the influence of various enzyme activity and the content of inflammatory factors reduced the damage degree of LPS,reached the purpose of protecting the liver and intestinal,the comprehensive utilization of fungus resude and deeply comorbidities of new drug research and development provided a new way of thinking,can effectively avoid waste,reduced the damage to the environment,so as to maximize efficiency. |
