Objective:Search CNKI,VIP,WanFang and Pubmed database to obtain the literature on the treatment of diabetes with urinary tract infection by Chinese herbal compound prescription.Through data mining,summarize modern Chinese medicine’s medication rules of the pathogenesis of diabetes with urinary tract infections and obtain core Chinese medicine combinations.Obtain the main active ingredients of the core Chinese medicine combination for the treatment of diabetes with urinary tract infections and their mechanism of action through network pharmacology research methods.Methods:The literature database was searched for the literature on the treatment of diabetes with urinary tract infection by Chinese herbal compound prescriptions,and the retrieved literature was screened according to inclusion and exclusion criteria.Sort out the prescription information included in the literature,and establish a database of Chinese medicine compound treatment for diabetes with urinary tract infection.After the entry is completed,the prescription information is standardized and imported to the ancient and modern medical record cloud platform.Explore the prescription of traditional Chinese medicine compound prescriptions for the treatment of diabetes with urinary tract infections through association rules,cluster analysis,complex network analysis and other methods,and obtain the core Chinese medicine combination.Search the core Chinese medicine combination through TCMSP to obtain the active ingredients and related targets of Chinese medicines,and obtain the corresponding gene names through the Uniprot database.Search the Gene Cards database and OMIM database for the target genes related to diabetes with urinary tract infection,and combine and remove duplicate values.Use Venny software to take the intersection of the obtained disease-related target genes and the target genes corresponding to the active ingredients of traditional Chinese medicine and obtain common target genes for diseases and core prescriptions.Use Cytoscape to draw the core prescription drug-component-disease-target gene and component-target gene network to screen the main active ingredients.Use the String database to construct protein-protein interaction network and screen key target genes.MCODE cluster analysis,GO function and KEGG pathway enrichment analysis are performed on the common target genes of traditional Chinese medicine diseases.Use Autodock software for molecular docking of key target genes and main effective ingredients.Results:The data mining results of the Chinese medicine compound treatment database for diabetes complicated with urinary tract infection showed that the commonly used Chinese medicines for the treatment of diabetes with urinary tract infection were Gancao,Fuling,Dihuang,Shiwei,Huangbai,etc.The most frequently used traditional Chinese medicine treatment is diuretic and Tonglin.In addition,it also includes clearing away heat and detoxifying,replenishing spleen and qi,nourishing yin and promoting fluid,expectorant,relieve pain and relieve pain,etc.Traditional Chinese medicine for the treatment of diabetes with urinary tract infections is mainly cold,slightly cold,mild,warm,and lukewarm,and the main medicinal taste is sweet,bitter,pungent,and mild.The meridians are mainly the lung,kidney,liver,Spleen meridian,heart meridian,stomach meridian and bladder meridian.The analysis of association rules of drug compatibility shows that the top three pairs of Chinese medicines with co-occurrence times are Zexie-Fuling,Bianxu-Qumai and Shanyao-Fuling.The results of cluster analysis of drugs with a frequency of medication ≥ 20 times finally resulted in 5 drug groups,including Fuling-Zexie-Danpi-Shanyao-Shanyurou,Gancao-Huashi-Qumai-Bianxu,Shiwei-Cheqiancao-Zhizi-Pugongying,Dihuang-Huangbai-Zhimu,Huangqi-Cheqianzi-Danggui-Baizhu-Dangshen-Baimaogen-Baihuasheshecao-Tufuling.The complex network analysis results show that seven medicines including Fuling,Gancao,Qumai,Shanyao,Zexie,Bianxu,and Cheqianzi are the core Chinese medicine combinations for the treatment of diabetes with urinary infections.The results of the network pharmacology study of the core Chinese medicine combination show that the active ingredients closely related to the target genes mainly include Quercetin,Kaempferol,Luteolin,Emodin,Naringenin and so on.Construct a protein-protein interaction network for common target genes,and screen key target genes including AKT1,IL6,TNF,TP53,MAPK3,etc.A molecular docking study of the main active ingredients and key target genes found that the binding energy of the main active ingredients and their target proteins were both less than-5kj/mol,suggesting that the two can bind stably.Conclusion:The analysis of the data mining results reflects the clinical drug rule of modern Chinese medicine for the treatment of diabetes with urinary tract infections and the understanding of it’s basic pathogenesis.And summed up the core traditional Chinese medicine combination consisting of seven medicines including Fuling,Gancau,Qumai,Shanyao,Zexie,Bianxu,and Cheqianzi.Analysis of active ingredients and target genes can infer that the core Chinese medicine combination obtained from data mining is mainly in AGE-RAGE signaling pathways in diabetes complications,MAPK signaling pathways,etc.,through the regulation of inflammation,metabolism,etc.Related factors regulate cell proliferation and apoptosis,play anti-inflammatory,anti-oxidant,regulate glucose and lipid metabolism,and anti-tumor effects to intervene and treat diabetes with urinary tract infections.
