| Purpose:With vosviewer visual analysis software,the TCM syndromes,pathogenesis of western medicine and the correlation between Chinese and western medicine treatment of Atherosclerosis(AS)were analyzed,providing theoretical basis for experimental research.On the other hand,the combination model of spleen qi deficiency in Apo E-/-mice with atherosclerosis was constructed to reveal that JPTMF can prevent and treat as early by alleviating the behavior,serum lipoprotein and inflammatory factor levels of as mice,and provide theoretical and experimental basis for clinical early prevention and treatment of atherosclerotic diseases.Material and method:1.Theoretical research:Search keywords such as"atherosclerosis""syndrome differentiation""syndrome elements""pathogenesis""inflammation""traditional Chinese medicine"and"compound prescription"in CNKI database,export the literature to refworks format,and make visual analysis with vosviewer software.2.Experimental study:Fifty 8-week-old SPF male Apo E-/-mice were randomly divided into5 groups:model group,JPTMF high-dose group,JPTMF medium-dose group,JPTMF low-dose group and atorvastatin group(hereinafter referred to as high-dose group,medium dose group,low-dose group and western medicine group),10 mice in each group were fed with high-fat diet,and 10 8-week-old SPF male C57BL/6J with the same genetic background were used as control group.The normal group was fed with normal diet.The model group and normal group were given the same amount of normal saline by gavage.The high-dose group,middle dose group,low-dose group and Western medicine group were given the same amount of JPTMF high-dose,middle-dose,low-dose and atorvastatin calcium tablets by gavage.The high-dose,middle-dose and low-dose ratio of JPTMF was 3:1:1/3.After one week of adaptive feeding,the rats were gavaged for 12 weeks,and the samples were taken at the end of 13 weeks.The weight and diet of the mice were measured once a week.The grip strength and anti fatigue ability of the mice were measured at the fourth,seventh,tenth and thirteenth weeks.In the 13th week,the field experiment,fecal moisture content and serum pancreatic amylase content of mice were determined.The contents of LDL,HDL,TC and TG in serum were detected by automatic biochemical analyzer.The expression levels of IL-4,IL-6,IL-10and TNF-αin serum were detected by enzyme-linked immunosorbent assay(ELISA).He staining and oil red O staining were used to observe the morphological changes and lipid deposition of aorta.Results:1 VOSviewe visualization analysis resultsThe key words of as closely related to TCM syndrome differentiation and syndrome elements are qi deficiency,phlegm turbidity,blood stasis and spleen deficiency.The commonly used treatment methods are promoting blood circulation and removing blood stasis,supplementing qi and activating blood circulation,etc.The key words closely related to the treatment of traditional Chinese medicine are Salvia miltiorrhiza,traditional Chinese medicine of promoting blood circulation and removing blood stasis;the commonly used prescriptions are compound salvia miltiorrhiza dropping pills,Xuefu Zhuyu Decoction,Buyang Huanwu Decoction,etc.In recent years,researches on as focus on chest obstruction,heart blood stasis,spleen deficiency and stroke.The key words closely related to pathogenesis are blood lipid,inflammation,vascular smooth muscle cells and so on.The key words closely related to inflammation are macrophage,oxidative stress,C-reactive protein,TNF-α,IL-6,IL-10,etc.,which are closely related to atorvastatin in treatment.2 Oil red O results showed thatCompared with the normal group,there was a large amount of lipid deposition in the aorta of the model group.Compared with the model group,the lipid status in aorta of each drug group was improved,especially in the high-dose group.3 HE staining results showed thatCompared with the normal group,the aorta structure of the model group was in disorder,the intima was not smooth,and there were foam cells in the middle wall of the tube,and there were obvious atheromatous plaques.Compared with the model group,the aortic structure of high-dose group,medium dose group and Western medicine group was significantly improved.4 Experimental resultsAfter 12 weeks of high-fat diet feeding,compared with the normal group,Apo E-/-mice in the model group showed obvious spleen qi deficiency related symptoms,such as decreased range of activity,grip,anti fatigue ability,diet and serum pancreatic amylase content(P<0.01),increased fecal moisture content and slow weight gain(P<0.01).At the same time,compared with the normal group,the serum HDL level of Apo E-/-mice in the model group decreased,LDL,TC and TG increased(P<0.01);the expression levels of serum anti-inflammatory factors IL-4 and IL-10 decreased,and the expression levels of pro-inflammatory factors IL-6 and TNF-αincreased(P<0.01).Compared with the model group,JPTMF could improve spleen qi deficiency and inflammatory reaction of Apo E-/-mice in a dose-dependent manner,and the effect of high-dose group was the best(P<0.01),and the effect was better than that of Western medicine group.Conclusion:1.The occurrence and development of atherosclerosis is closely related to the function of spleen.The treatment of atherosclerosis by invigorating the spleen,replenishing qi and activating blood circulation is effective.2.JPTMF can improve the blood lipid level of Apo E-/-mice by increasing the content of HDL and reducing the contents of LDL,TC and TG.3.The mechanism of JPTMF in the treatment of as is to improve the inflammatory reaction in vivo of Apo E-/-mice,increase the levels of anti-inflammatory factors IL-4 and IL-10 in serum of Apo E-/-mice,and inhibit pro-inflammatory factors IL-6 and TNF-αIt’s related to the level of education. |
