Study On Starch-Ganoderma Lucidum Triterpenoid Ball Milling Complex For Relieving Body Damage Under Stress State

Triterpenoids have various biological activities such as anti-tumor,anti-inflammatory,immune regulation and liver protection.Cyclophosphamide(CP)is a widely used anti-tumor drug in clinical practice.Its shortcomings are lack of tumor cell specificity,toxic side effects on the immune system and multiple organs of the body.this paper takes starch-Ganoderma lucidum tri terpenoid ball milling complex as the research object,and discusses its regulation effect on the side effects of CP-induced rats.METHODS:SD rats were divided into normal control(NC)group,model control(MC)group,starch-Ganoderma lucidum triterpenoid simple mixture+CP(MTC)group,starchGanoderma lucidum triterpenoid ball milling compound+CP(CTC)group,starchGanoderma lucidum triterpenoid simple mixture(MT)group and starch-Ganoderma lucidum triterpenoid ball milling compound(CT)group,continuous gavage for 7 days.On the 7th day,rats in the MC,MTC and CTC groups were intraperitoneally injected with 200 mg/kg body weight CP.The effects of the interventions on the immunodeficiency,hepatotoxicity and changes in intestinal flora of CP-induced rats were studied respectively.RESULTS:①Ganoderma lucidum triterpenoid can slow down the weight loss of rats caused by CP and increase the immune organ index.Ball milling complex intervention significantly improved the spleen index of rats(P<0.05).Ganoderma lucidum triterpenoid intervention had no significant effects on the content of cytokines and immunoglobulins and expression of inflammation-related genes in rats.Ganoderma lucidum triterpenoid can significantly improve the related indexes of antioxidant defense function in serum and spleen tissue.Compared with MC group,the levels of SOD in serum and GSH in spleen homogenate in CTC group increased significantly(P<0.05).Meanwhile,Ball milling compound can increase the mRNA expression levels of SOD3,GPX2,Nrf-2,HO-1 genes in spleen tissue,so as to resist the production of oxidative stress caused by CP injection.②Ganoderma lucidum triterpenoid can improve the elevation of liver function marker enzymes in serum induced by CP and reduce the pathological changes of liver tissue to some extent.The ball milling complex significantly decreased the elevation of serum ALT(P<0.05).Both interventions significantly reduced MDA content in liver homogenate(P<0.05)and increased levels of GSH,T-AOC and CAT.③Ganoderma lucidum triterpenoid can improve the abundance and diversity of rat intestinal flora.Compared with the MC group,the abundance of Deltaprotobacteria in the MTC and CTC groups decreased,while that of Bacteroides acidifaciens,Bifidobacterium pseudolongum,Alloprevotella,Ruminococcus,Prevotella and Lactobacillus increased.
