The polysaccharide and flavonoid saponin of Radix Polygonatum were extracted from Radix Polygonatum Polygonatum as raw materials to determine the best extraction scheme of the total functional components of Radix Polygonatum.And the yellow essence extract as raw material to ethanol mixed with edible yellow base,validation solomonseal base effect of antioxidation and anti-fatigue.Finally,the extract of Rhizoma Polygonatum was used as the main raw material to prepare Rhizoma Polygonatum liquor according to the national standard of liquor preparation,and the following conclusions were drawn:(1)To determine the best extraction conditions for the effective components of Radix Polygonatum Radix as raw materials by conducting extraction experiments on the effective components of Radix Polygonatum Radix.The optimal extraction conditions were as follows:the mass ratio of Radix Polygonatum to ethanol volume(g/mL)was 1:20,the extraction temperature of ethanol water bath was 50℃ the concentration of ethanol was 30%,and the extraction time of ethanol water bath was 3 h.The yield of polysaccharide was 8.71%±0.13%.The optimal extraction conditions for flavonoids were as follows:the material ratio(g/mL)of C.flavescens to ethanol was 1:20,the temperature of ethanol water bath was 50℃,the concentration of ethanol was 50%,and the extraction time was 5 h.The yield of flavonoids was 6.84%±0.525%,The optimal extraction conditions were as follows:the ratio of ethanol to rhizoma flavescens(g/mL)was 1:20,the water extraction temperature of ethanol was 50℃,the concentration of ethanol was 50%,and the extraction time was 4 h.The yield of xanthosaponin was 6.71%±0.326%.The total functional components were extracted by stepby-step extraction,and the material ratio of fixed Rhizoma cymbidii and ethanol was 1:20(g/mL).The optimal conditions were as follows:ethanol water bath extraction temperature was 50℃,ethanol concentration was 30%,and ethanol water bath extraction time was 3 h.The extraction temperature was 50℃,the concentration of ethanol was 50%,and the extraction time was 4h in ethanol water bath.The extraction temperature of ethanol water bath was 50℃,the concentration of ethanol was 50%,and the extraction time was 5h.After three times of extraction of the total functional components of Radix Polygonatum,the final yield of polysaccharide,flavonoids and saponins was 9.36%,7.82%and 7.43%.(2)The efficacy of Huangjing based liquor prepared with the functional components of Huangjing and edible ethanol was evaluated,and the anti-oxidation and anti-fatigue effects of Huangjing on mice were evaluated.The results showed that huangjingji liquor could increase the activity of catalase,glutathione content,superoxide dismutase activity,and inhibit the production of malondialdehyde in mice,and had the corresponding antioxidant activity.At the same time,it can prolong the exhaustion time of mice,increase the glycogen reserve in mice,and reduce the content of lactic acid,which has a positive effect on anti-fatigue.In the detection of alcohol liver injury,base.liquor had little effect on the activity of alanine aminotransferase in mice,and little effect on liver.(3)Polygonatum liquor was prepared with the functional ingredients of yellow essence liquor as raw material.When the ratio of yellow essence base liquor and Maotai-flavor liquor was 1:10,the yellow essence liquor had the best taste and the liquor body was clear.Extreme conditions and stability tests had no effect on the liquor body.The indexes of Huangjing liquor meet the national standard.
