Objective To compare the structure and diversity of intestinal flora between ulcerative colitis(UC)patients and UC patients with nonalcoholic fatty liver disease(NAFLD)and healthy people,and to explore the characteristics of intestinal flora in UC patients with NAFLD.Methods According to the inclusion criteria,5 cases of UC patients with NAFLD and 22 cases of UC were selected as control group,8 cases of UC and 9 cases of healthy people as control.Collection of fresh feces naturally discharged by selected persons.The peripheral ice bags were transported to the laboratory of ** and then stored in the refrigerator at-80℃.The enteric microflora DNA was extracted from Hangzhou Jingbai Technology Co.,Ltd.High throughput sequencing of the 16 S r DNA V3-V4 region was carried OTU by Illumina Miseq platform.The structure and diversity of flora abundance were obtained.Results In UC patients with NAFLD group,the bacteria with higher abundance in phylum,class,order,family,genus were Verrucomicrobia,Verrucomicrobiae,Lachnospiraceae,Roseburia and Pseudomonadales,Pseudomonadales,Lactobacillaceae,which were lower than those in UC group.The difference was statistically significant(P<0.05).There was no significant difference in flora structure between UC group and healthy control group at phylum level.At the class level,there were 3 species of fungi in the two groups.At the family level,there were 1 species of bacteria with significant difference,that is,the abundance of Acidaminococcaceae in UC group was significantly lower than that in healthy group(P<0.01).At the genus level,the abundance of Clostridium XlVa(P<0.05)and Phascolarctobacterium(P<0.01)decreased in UC group.Conclusion 1.Pseudomonadales and enterococcaceae in UC patients with NAFLD was lower than that in UC patients.Lachnospiraceae and roseburia in UC patients with NAFLD was higher than that in UC patients.But the number of lactobacillaceae is down,which brings inspiration to the clinical treatment of patients with complications in the future.2.The number of intestinal beneficial bacteria in UC patients was lower than that in healthy people,and the specific flora was lower than that in healthy people and more than that in UC patients with NAFLD.
