| Background The processing of producing areas is a series of rough processing in the producing areas before the chinese herbal medicines are imported into the decoction pieces companies.There are still many problems in the processing of producing areas.The processing of producing areas is based on traditional processing,processing methods are not uniform,and the technology and equipment are large,and the technology and equipment are backward and can not be produced in a large scale.The processing of producing areas is based on small workshops,scattered and confused,which leads to the difficulty of supervision,resulting in adulteration,dyeing and sulfur in the processing of producing areas.Therefore,in view of the above problems,the State Administration of traditional Chinese medicine has initiated the research on the integrated specification technology of fresh cut processing of Chinese medicinal materials.The laboratory has undertaken the task of the Radix Linderae.Objective 1.To Establishment of evaluation standard for fresh-cut processing technology of Radix Linderae in origin,in order to provide theoretical basis for the standardized production of Radix Linderae.2.To Establish fresh-cut processing technology of Radix Linderae in origin,in order to guide the standardized production of Radix Linderae.3.To evaluate the equivalence of fresh-cut Radix Linderae pieces in origin and traditional processed pieces,Verify whether there is a difference between the two processing methods.Methods 1.To examine the traits,identification,moisture content,total ash content,sulphur dioxide residue,leachate content,and contents of main active ingredients of24 batches of Radix Linderae Pieces and 4 batches of fresh-cut pieces in origin,and to develop a more demanding standard based on the current pharmacopoeia standards to improve the quality of Radix Linderae pieces.The content was determined by high performance liquid chromatography.2.The effect of five different drying methods on the fresh cutting of the producing area of the black medicine was investigated by the evaluation standard of the Chinese medicinal herbs,and the drying method suitable for the fresh cutting of the producing area was selected.Taking the hot air drying temperature,loading capacity and slice thickness as the inspection factors,the changes of water content during the drying process were measured in real time.The curve of the moisture ratio and drying rate were plotted.By fitting nine classic thin-layer drying models,the model which suitable for the drying of Radix Linderae was selected and the parameters were solved to obtain a model that can guide the drying of Radix Linderae in real time.Taking the content of leachable extracts,Linderane and Norisoboldine as optimization indicators and taking slice thickness,drying temperature,and dry Loading capacity as the inspection factors for fresh-cut processing of Radix Linderae in origin,In response to the three factors,we optimized the response surface to obtain an optimal processing parameter for the processing area of Chinese herbal medicine.3.To compared the physicochemical traits of fresh-cut and traditional processed Radix Linderae through the established criteria for the assessment of Radix Linderae pieces to verify that the fresh-cut processing of Radix Linderae in origin are non-inferior to the traditional processed Radix Linderae in physical and chemical properties.To establish acetic acid writhing analgesic model,hot plate analgesia model and estrogen-induced dysmenorrhea model,record the writhing response,hot plate jump reaction,and the changes of prostaglandin E2(PGE2),prostaglandin F2α(PGF2α),vasopressin(AVP),andβ-endorphin(β-EP)to compare the bioequivalence of pain relief with two different processing methods.Results 1.28 batches of samples were all in accordance with the trait identification,TLC identification,and microscopic identification characteristics of Radix Linderae,and the difference between these samples were not great;The water standard of 28batches of Radix Linderae was 10%;The total ash standard is 4%;The acid-insoluble ash standard is 2%;The SO2 residue standard is no more than 140 ppm;The leachate content standard is 12%.The Linderane content standard is 0.070%.The Norisoboldine content standard is 0.4%;The Linderalactone content standard is0.025%.The lindenenol content standard is 0.010%.2.There was no significant difference between stoving and other four drying methods from the indicators of traits,identification,content of leachables,content of main chemical components,while moisture and total ash were superior to the other four drying methods.In general,stoving was not inferior to the other four drying methods.The Page model was the best model(R2>0.996)for the hot air drying of Radix Linderae.When the temperature was between 40°C and 90°C,the effective diffusion coefficient were ranges from 3.2018×10-9 to 9.8890×10-9 m2/s and the dry activation energy is 26.32 k J/mol;The optimized results of response surface were dry loading of8m2/kg,drying temperature of 65°C,slice thickness of 2mm,The result was in line with the facts andthe optimization result was desirable.3.There were no significant differences in the characters,microscopic identification and TLC identification of fresh and processed Lour and fresh Lour.They all conform to the characteristics of the Radix Linderae.The results of examination and content determination of two kinds of processed products were in line with the newly established standards,and there was no significant difference between them.In three animal models,compared with the model group,the two different methods of Radix Linderae processing can both significantly reduce the writhing frequency(P<0.01),and significantly prolong the thermal plate pain threshold time(P<0.05),decreased PGE2,PGF2α,AVP content(P<0.05),increasedβ-EP content(P<0.01),but there was no significant difference at the same dose(P>0.05).Conclusion 1.The evaluation criteria established for fresh-cut processing technology of Radix Linderae in origin has been improved on the basis of pharmacopoeia,and it can well control the quality of the fresh-cut slices of Radix Linderae in origin.2.Stoving method was selected as drying method for fresh-cut processed Radix Linderae in origin.The established drying kinetic model can objectively reflect the drying process of Radix Linderae,and the processing parameters of fresh-cut processing obtained by the response surface optimization method was feasible,which can provide a reliable theoretical basis for the production and processing of Radix Linderae pieces.3.There was no significant difference in quality and efficacy between fresh and fresh processed Chinese herbal medicine and traditional Chinese medicine. |
