Objective:1.Through the investigation of the production sites of Paeoniae Radix Alba in Bozhou,we can confirm the germplasm of Paeoniae Radix Alba in Bozhou in order to determine the origin of the certified products of Paeoniae Radix Alba in Bozhou,and to guide the correct selection of the germplasm in the production area of Paeoniae Radix Alba in Bozhou.2.To investigate the characteristics and quality of the common cultivated products of Paeoniae Radix Alba in Bozhou,and to select the cultivated varieties with high quality and high yield,and to clarify the importance of the purified varieties.3.Formulate the standardized planting operation procedures to improve the quality of Paeoniae Radix Alba in Bozhou.4.Explore the best slice thickness of Paeoniae Radix Alba in Bozhou,and explore the scientific connotation of the production site to cut Paeoniae Radix Alba in Bozhou into thick slices.5.To study the feasibility of the application of integration of primary processing and processing technology of Paeoniae Radix Alba in Bozhou and explore the quality of quality of slices by integration of primary processing and processing technology,which made the processing of Paeoniae Radix Alba in Bozhou slices more time and labor-saving and improved the quality of Paeoniae Radix Alba in Bozhou.Methods:1.Using HPLC fingerprint and the simultaneous assay method for gallic acid,oxidation paeoniflorin,albiflorin,paeoniflorin and benzoic acid,1,2,3,4,6-O-five gallic acid and glucose and benzoypaeoniflorinl to study traditional bleeding bud of Paeoniae Radix Alba in Bozhou and seed breeding of paeonia lactiflora and to cluster analysis seven kinds of component content.2.Take a summary of the research on the characteristics and quality of different cultivated species of Paeoniae Radix Alba in Bozhou through field investigation and literature review.3.Through on-site tracking of the selection of land,site preparation,preparation of buds,cultivation and management,and other links,we visited farmers,and summarized the standardized planting procedures for Paeoniae Radix Alba in Bozhou.4.Through the analysis of the detrital rate,off-cut rate and warping rate of the pieces cutting of 1.5,2.2,3.0,and 3.5 mm in thickness of Paeoniae Radix Alba in Bozhou,dry extract yield rate of different thickness pieces,and the decocting rate of six ingredients,such as galic acid,determine the thickness of the appropriate cut combining with the loss rate and quality.5.By processing 10 batches of fresh products of Paeoniae Radix Alba in Bozhou from several local townships in Bozhou,the traditional methods of processing were used to prepare the paeioniae radix alba and the traditional boiled slices and the freshed boiled slices and the raw oblique slices which are prepared by processing of origin and processing.And then determine and compare the similarity of fingerprints with different slices of different processing mode of Paeoniae Radix Alba and seven components of Paeoniae Radix Alba,and compare the fingerprint of the traditional boiled slices,fresh boiled slices and fresh unboiled slices,the content and fingerprint of seven components of paeoniar radix alba and the common peaks of 30 batches of Paeoniae Radix Alba pieces were clustered.Results:1.The method of determination of the fingerprint and multicomponent content of the radix paeoniae and its standard decoction was well established.2.There was a great difference in the germplasm quality of the Paeoniae Radix Alba in Bozhou,and the similarity degree of the diversification of the diploids was significantly different from that of the traditional Paeoniae Radix Alba in Bozhou.The peony alkaloids,paeoniflorin,and benzoyl glycosides were found in the hybrids.The total glucosides of peony(the sum of the four monoterpene glycosides)and the traditional were compared,P<0.01,with very significant difference;cluster analysis of the content of the components in the different germplasms of the white peony combined the traditional pupa white pupa into a class,mixed with white flowers are clustered into one category.3.The characteristics of different rhiaomes,such as“xiantiao”,“pubang”,“jizhua”,“mazha” are different,and the main components in different cultivations are different.The more common varieties are also different in kinship,and the growth years of different cultivated variety are also different.The phenomenon of mixed cultivation of multiple crops has reduced the quality of white peony.4.The standardized planting operation procedures were established by investigation.5.The detrital rate,off-cut rate,warping rate,and the dry extract yield rate of Paeoniae Radix Alba in Bozhou with different thicknesses,and the decocting rate of six ingredients,such as gallic acid,etc.,were as follows: as the thickness of the decoction pieces increased,the detrital rate,off-cut rate and the dry extract yield was gradually increased,and the extract efficiency of the main components was slightly lower.However,the thickness of the decoction pieces was from 1.5mm to 2.2mm,and the change rate of the six components was not significant(P>0.05),while the thickness of the decoction pieces was from 3mm to 3.5mm,the paeoniflorin frying rate was significantly reduced(P <0.05).6.The research on the integration of production processing and processing in the production area of Paeoniae Radix Alba in Bozhou showed that the similarity difference between the fingerprints of Paeoniae Radix Alba in Bozhou and other processing methods was compared with that of the raw oblique group,and that of the traditional slices,fresh boiled slices,and Paeoniae Radix Alba in Bozhou P<0.05.The similarity between the traditional boiled slices and Paeoniae Radix Alba in Bozhou was compared,P<0.05.The similarity degree of the standard decoction of different processing methods was similar to that of the traditional boiled slices.The fresh tablets and the traditional slices had P<0.05;The results of the determination of various components in the processing method decoction pieces showed that the content of paeoniflorin in Paeoniae Radix Alba in Bozhou was significantly higher than that of traditional slices(P<0.05)and the content of benzoic acid was significantly lower than that of traditional slices(P<0.05);The content of paeoniflorin and paeoniflorin in raw oblique slices was significantly higher than that of traditional slices(P<0.05),1,2,3,4,6-O-pentagluacylglucosamine content was significantly lower than that of traditional slices(P<0.05);different processing methods In the study of the standard decoction of Paeoniae Radix Alba,the content of gallic acid,paeoniflorin,and benzoic acid in raw rumas slices was higher than that of the traditional slices.The content of benzoic acid in the raw rumas slices was higher than that of the two boiled slices.The content of 1,2,3,4,6-O-pentamorphoyl glucose snd Benzoyl paeoniflorin was lower than that of the two cooking products.The transfer rate of major components in different decoction pieces was that the transfer rate of paeoniflorin,paeoniflorin,and benzoic acid components in traditional slices was significantly different from that of slices(P<0.05),and the transfer of paeoniflorin and paeoniflorin from traditional tablets was significant.The rate was higher than that of fresh slices,and the transfer rate of benzoic acid considered to be a harmful ingredient was higher than that of traditional slices.There was no significant difference between the other ingredients in the two ingredients;four monoterpene glycosides in the Paeoniae Radix Alba raw rumas slices.The transfer rate of pentagalactosyl glucose and benzoic acid components was significantly lower than that of traditional slices(P<0.05).Conclusion:1.The establishment of Paeoniae Radix Alba in Bozhou and its standard decoction fingerprint and multi-component content determination method is stable and reliable,which can be more comprehensive evaluation of the quality of radix paeoniae.2.The fingerprints of Paeonia lactiflora,which were propagated by seeds,had low similarity with traditional Paeonia lactiflora Pall.The contents of different components were quite different,and the germplasm quality difference was great.It should not be applied to the market with traditional paeony and applied to clinic.3.Cultivars of Paeoniae Radix Alba rhiaomes mixed in planting,with different germplasm and different growth years,which was affecting the quality and yield of Paeoniae Radix Alba.So we should purifying the germplasm of Paeoniae Radix Alba vigorously.4.The results of the study on the loss rate and the percentage of extractives of the main component of different slices of Paeoniae Radix Alba in Bozhou show that the traditional cutting method of Paeoniae Radix Alba are not very reasonable for1~2mm slices,and 2~3mm thick slices not only can significantly reduce the loss,but also can not result in the decreasing rate of effective substance decoting.It is suggested that the thickness of Paeoniae Radix Alba slices should be below 3 mm,that is,no more than 3 mm.5.There is no significant difference in the quality of fresh boiled slices of Paeoniae Radix Alba,which is produced by integrated process of processing and processing in the production area,which can save time and energy and promote the non-vulcanization process of paeoniae.
