Objective: The gene chip technology was used to detect the m RNA expression levels of TLR/My D88 signaling pathway-related genes in the kidney tissues of patients with idiopathic membranous nephropathy and the control group,so as to screen for differentially expressed genes.Methods: Collect five patients with membranous nephropathy diagnosed by renal biopsy in the Second Affiliated Hospital of Guangxi Medical University as case group.In the control group,there were 3 patients who underwent nephrectomy for renal cancer in the urology department of our hospital.Kidney tissues with no abnormal pathological results were taken away from the cancer tissue by more than 5cm.Total RNA was extracted from kidney tissues of five cases in the case group and three cases in the control group,and then reverse transcribed into c DNA.Gene chip technology was used to detect the m RNA expression of genes related to the TLR/My D88 signaling pathway.Statistical analysis was performed using SPSS 22.0 software,and P<0.05 and the gene expression level was up-regulated or down-regulated by more than 1.5times as the difference was judged to be statistically significant.Perform GO function analysis and KEGG pathway analysis of differential genes,as well as analysis of interactions between genes encoded proteins.Results: Among the 84 genes related to the TLR/My D88 signaling pathway,CD80,IFNB1,IL1 A,IL6,and IL17 A were not further analyzed because the expression of the 5 genes was too low.Compared with the control group,among the remaining 79 genes,the expression of 7 genes in the idiopathic membranous nephropathy group was different.Among them,there was 2 up-regulated gene,they are CD14(2.56 times,P=0.038633)and TLR6(4.57 times,P=0.026339).There are 5 down-regulated genes,respectively are IFNA1(-5.21 times,P=0.000182),JUN(-2.2 times,P=0.048926),NFKBIA(-1.72 times,P=0.047605),SIGIRR(-1.55 times,P= 0.01936),TICAM1(-3.07 times,P=0.03716).GO analysis showed that differentially expressed genes are mainly involved in lipopolysaccharide-mediated signaling pathways,innate immune response,My D88-dependent and non-dependent toll-like receptor signaling pathways and other biological processes,mainly enriched in Toll-like receptor signaling pathways,and there are interactions between proteins.Conclusion: This study found that a total of 7 genes were abnormally expressed in 84 TLR/My D88 signaling pathway-related genes in the kidney tissue of patients with Idiopathic membranous nephropathy.These differentially expressed genes mainly involve toll-like receptors,adaptor protein gene,toll-like receptor negative regulatory molecule,downstream genes of the TLR signaling pathway,and inflammatory cytokines,suggesting that the TLR signaling pathway may be an important immune regulatory signaling pathway leading to the pathogenesis of idiopathic membranous nephropathy. |
