Objective: The thesis aimed to investigate the physiological indicators, intestinal flora and taste differences in three different constitutions in Traditional Mongolian Medicine(TMM)and to clarify the scientific connotation of the correlation between basic taste and digestive taste and TMM body constitutions,so as to provide a basis for personalized drug use in the clinics. Methods: According to the Mongolian Medicine Constitution Classification and Evaluation Table and Mongolian medicine diagnosis method, groups with different Mongolian medicine body constitutions were determined, basic physiological indicators were measured, urine and saliva were collected for further analysis.Fecal samples were collected and llumina Nova Seq sequencing method was used to test the fecal flora of people with different constitutions.According to the species annotation, the Alpha diversity and Beta diversity as well as the differences between groups were analyzed to explain the differences between constitutions. The concentration gradients of six basic Mongolian medicine tastants sour, sweet, salty, astringent, spicy and bitter were selected to observe the perception threshold and identification threshold of different taste perception of people with different constitutions. Results: A total number of 283 subjects was conduced the questionnaire and the response rate was 100%.After the questionnaire classification,24 subjects had Heyi constitution,57 subjects had Xila constitution,13 subjects had Badagan constitution,42 subjects had Mixed constitution, and the remaining 147 subjects were others.Physiological indexes: there was no significant difference among Heyi group,Xila group,Badagan group and Mixed group. There were significant differences in BMI between badagan group and Heyi group(P<0.05),between Badagan group and Xila group(P<0.05),and between Badagan group and Mixed group(P<0.05).There was significant difference in saliva flow rate between Mixed group and Heyi constitution group(P<0.05).The difference in Bacteroidota in Heyi constitution group was significantly higher than that in Mixed constitution group(P<0.05),while there was no significant difference in Xila constitution group,Badagan constitution group and Mixed constitution group. The Proteobacteria in Badagan constitution group was significantly higher than that in Heyi constitution group(P<0.05),but there was no significant difference among other groups. There was statistical significance in Megamonas between Heyi group and Badagan group(P<0.05).There were significant differences in Prevotella and Bacteroides between Xila group and Badagan group(P<0.05).There was statistically significant difference in Bacteroides between Xila group and Mixed group(P<0.01).The relative abundance of Faecalibacterium in badagan group was significantly higher than that in Mixed group(P<0.05).A significant difference in detection threshold was found in astringency between badagan and Mixed group(P<0.05)a different recognition threshold in saltiness between Xila and Badagan group(P<0.05).Conclusion: This study showed that there were significant differences in body weight, salivary flow rate and intestinal microbial structure among heyi,Xila,Badagan and Mixed constitutions, but no significant differences in urine indexes were found. In addition, significant differences in the perception of saltiness and astringency among people with four different constitutions were found, which is probably related to different dietary habits and gastrointestinal microbial structure. The correlation of different microbial structures and taste perception between different constitutions needs further study.Overall,by analyzing the oral taste perception and gastrointestinal microbial structure of different constitutions of Mongolian medicine, this paper preliminarily explored the internal relationship between constitution-basic flavor and digestive flavor of Mongolian medicine, laying a foundation for the scientific interpretation of individualized drug use of different constitutions of Mongolian medicine.
