Association Between CD14 Promoter Gene-159(C-T) Polymorphism And Foot Dorsal Arteriosclerosis In Diabetic Feet Patients

Objective:To study the association between polymorphism of the CD14 gene at the159 locus and foot dorsal arteriosclerosis(FDA)in Chinese Korean Type 2 diabetic feet(DF)patients,exploreing the risk factors and providing clinical warning information for Korean DF patients in Yanbian area.Methods:A total of 286 consecutive patients of Korean nationality with diagnosed DF in our hospital from October 2018 to May 2020 were enrolled in this research.They were all examined by color doppler ultrasound for foot dorsal arteriosclerosis,then the DF patients with arteriosclerosise(FDA)were divided into 216 without ulcer(NFDAU)and 70 ulcer group(FDAU).The blood biochemical indexes of patients were recorded.CD14 promoter gene-159(C-T)polymorphism was analyzed by restriction fragment length polymorphism(RFPL).The age,sex,smoking and alcohol consumption of all patients were recorded in detail.According to the standardization recommendations of WHO,the definition of smoking was"smoking>4 times per week,with a total of≥6 cigarettes".Drinkers were defined as"average intake of alcohol>30 g/day and duration≥6 months".Blood pressure,course of disease,body mass index(BMI),blood pressure,blood glucose,blood lipids,low density lipoprotein and creatinine were measured.Results:There was no significantly difference between genotypes and alleles in CD14 promoter-159 gene(x~2=1.801,P>0.05;x~2=1.864,P>0.05).The distributions of CC genotype vs CT+TT genotype had significantly difference in CD14 promoter-159 gene between Korean FDA patients without or with ulcers(x~2=0.009,P<0.05).There were significant differences between NFDAU and FDAU groups in disease course,smoking and drinking,hyperglycemia and hyperuric acid(P<0.05).Logistic regression analysis showed that disease course,smoking and alcohol consumption,and C allele were associated with FDAU(P<0.05).Conclusion:(1)There is relation between distribution of CD14 promoter gene-159(C-T)polymorphism with Korean FDA without or with ulcer.(2)The CC genotype is a susceptibility gene for the Korean FDA patients with ulcer.(3)Disease course,smoking,alcohol consumption and C allele were risk factors for FDAU.(4)CDUS can provide a variety of reliable indicators and information for DF patients.
