Objective: To observe the effect of ginger separated moxibustion at Shenque point on behavior and gastrointestinal motility in rats with functional gastrointestinal diseases of liver depression and spleen deficiency(FGIDs);The effect of ginger separated moxibustion on adrenergic receptor was observed from three targets of brain,gastrointestinal smooth muscle and vascular smooth muscle α2(α2-AR),vasoactive peptide(VIP)and its receptor VIPR1.Compared with mosapride and Prozac,to explore the molecular mechanism of ginger separated moxibustion promoting gastrointestinal motility and antidepressant through brain smooth muscle axis.Methods: 50 male SPF SD rats were randomly divided into normal group(group A),model group(group B),ginger separated moxibustion group(Group C),Mosapride group(Group D)and Prozac group(Group E).Group A was fed normally,and the other four groups were modeled by compound etiology method for 21 days.After successful modeling,group a continued to feed normally;Group C was bound and bound,moxibustion at Shenque point with ginger separated for 30 minutes,and perfused with 0.9%sodium chloride solution;The other three groups were bound for 30 minutes every day;Group B was perfused with 0.9% sodium chloride solution;Group D was perfused with Mosapride solution;Group E was given Prozac solution for 14 days.The general changes of rats in each group were observed.Open field experiments were conducted before and after modeling and after intervention;After the experiment,systematic anatomy was performed to determine the gastric residual rate and small intestinal propulsion rate;Western blot was used to detect the contents of smooth muscle in brain,stomach,small intestine and vascular smooth muscle of rats in each group α2-AR,VIP and their receptor VIPR1 protein expression.Result:1.General changes of rats: before the experiment,the rats in each group were in good spirits,sensitive and lively,clean and bright fur,clean eyes,pink auricles and spherical feces.After the completion of modeling,the general state of rats in group A was the same as before.The rats in other groups were tired and sleepy,with slow reaction and action,withered and yellow hair,light auricle color,pale eyes,more secretions,and dry and thin stools.After the intervention,compared with the rest groups,the rats in group C had better spirit,increased mobility,neat fur,no secretion in cleft eyes,light red auricles and moderate dry and sparse feces.There was no significant change in group B,group D and group E.2.Results of open field experiment: there was no significant difference in the number of climbing lattice,standing and modification of rats in each group before making the dog,which was comparable among the groups.After modeling,compared with group A and before modeling,the number of climbing lattice,standing and modification of rats in other groups decreased significantly(P<0.01).It shows that modeling can significantly reduce the number of grid climbing,standing and modification of rats.After the intervention,group A was relatively stable compared with that after modeling;In group C,the number of climbing grids increased significantly(P<0.01),the number of standing and modification increased,and the difference was statistically significant(P<0.05);There were no significant changes in other groups.It shows that ginger separated moxibustion can increase the number of grid climbing,standing and modification of rats.3.Changes of gastric residual rate and small intestinal pushing rate in rats: after the intervention,compared with group A,the small intestinal pushing rate in group B and group D decreased and the gastric residual rate increased,the difference was statistically significant(P<0.01).Compared with group B,the small intestinal propulsion rate in group C and group E was significantly higher(P<0.05),and the gastric emptying rate was significantly lower(P<0.01).4.In hypothalamus,stomach,small intestine and abdominal aorta of rats α2-AR;after the intervention,compared with group A,there were significant differences in hypothalamus,stomach,small intestine and abdominal aorta in group B,the expression levels of α2-AR decreased significantly(P<0.01);Compared with group B,hypothalamus,stomach,small intestine and abdominal aorta in group C,group D and group E,The expression level of α2-AR increased significantly(P < 0.01).5.Protein expression of VIP in hypothalamus,stomach,small intestine and abdominal aorta of rats: after the intervention,compared with group A,the expression level of VIP in hypothalamus,stomach,small intestine and abdominal aorta of rats in group B increased significantly(P<0.01);Compared with group B,the expression level of VIP in hypothalamus,stomach,small intestine and abdominal aorta of group C,group D and group E decreased significantly(P<0.01).6.Protein expression of VIP receptor vipr1 in hypothalamus,stomach,small intestine and abdominal aorta of rats: after the intervention,compared with group A,the expression level of vipr1 in hypothalamus,stomach,small intestine and abdominal aorta of rats in group B increased significantly(P<0.01);Compared with group B,the expression level of VIPR1 in hypothalamus,stomach,small intestine and abdominal aorta of group C,group D and group E decreased significantly(P<0.01).Conclusion:1.The rat model of functional gastrointestinal disease with liver depression and spleen deficiency was successfully reproduced.2.Ginger separated moxibustion at Shenque point can significantly increase the number of grid climbing,standing and modification of FGIDs rats in the open field experiment,and can also significantly promote the emptying of stomach and the advancement of small intestine in FGIDs rats.Its effect is basically equivalent to that of Mosapride,indicating that ginger separated moxibustion can not only resist depression,but also promote gastrointestinal motility.3.Ginger separated moxibustion at Shenque point can significantly improve the levels of gastrointestinal,hypothalamus and abdominal aorta in FGIDs rats with liver depression and spleen deficiency α2-AR protein expression,down regulating the protein expression of VIP and its receptor VIPR1,indicating that brain smooth muscle axis is an important way of ginger separated moxibustion.
