| Objective: To investigate the mechanism of Zhenganxifeng Decoction in treating Essential Hypertension on the basis of Network Pharmacology,and to evaluate the clinical efficacy of Zhenganxifeng Decoction in the treatment of Essential Hypertension(grade 1)with Yin eficiency and Yang hyperactivity syndrome,it provides a new research perspective and intervention measures for the prevention and treatment of Yin deficiency and Yang hyperactivity hypertension.Method: 1.Network Pharmacology Research: Through TCMSP database to screen out the effective components and action targets of Zhenganxifeng Decoction,through TTD,Drug Bank,Gene Card and other databases to mine the action targets of Essential Hypertension,through Venny diagram to get the common targets of both,by Cytoscape3.7.2,the target network of Traditional Chinese Medicine disease-component-target was plotted,and the interaction network of key targets was established by using STRING database.At last,the key targets were analyzed by GO and KEGG pathway.2.Clinical Study: 60 patients with Yin deficiency and Yang hyperactivity syndrome(grade 1)were divided into the Experimental Group(N = 30)and the Control Group(N = 30)according to the method of random number table.The experimental group was given the granule of Zhenganxifeng Decoction,the Control Group was treated with Valsartan capsule(Daiwen)for 28 days to evaluate the antihypertensive effect,TCM Syndrome and the improvement of quality of life,metabolism and safety.3.Target Verification:to observe the effect of Zhenganxifeng Decoction on PI3 K,AKT1 and IL6 in P13K/ AKT signaling pathway by collecting the serum of Experimental Group,Control Group and healthy volunteers before and after treatment.Result: 1.Network Pharmacology Research: 110 Chemical Constituents and1585 prediction targets of Zhenganxifeng Decoction were obtained by searching the database of TCMSP,and a total of 259 action targets were removed,according to the diseas gene database,5474 Essential Hypertension related targets were identified,and 83 common targets were identified,including PTGS2,NCOA2,DPP 4,NOS2,AR and so on.GO and KEGG were analyzed by David Database,and 555 biological processes and 127 signal pathways were obtained.2.Clinical Study: Zhenganxifeng Decoction can reduce blood pressure(p<0.05),the effect of lowering blood pressure is similar to that of valsartan capsule(p>0.05),it can improve TCM syndrome and quality of life(p<0.05),and the effect is better than Valsartan capsule(p<0.05),it has a certain curative effect(p<0.05)to improve the blood lipid and uric acid of the patients,and the curative effect is similar to that of valsartan capsule(p>0.05).There is no abnormal function of liver and kidney in the Experimental Group and the Control Group.3.Target Verification:Before treatment,serum PI3 K,AKT1 and IL6 in the Experimental Group and the Control Group were higher than those in the healthy volunteers.After 28 days of treatment,the expression of PI3 K,AKT and IL6 in the Experimental Group and the Control Group decreased,and the trend of decrease in the Experimental Group was not higher than that in the Control Group(p>0.05).Conclusion: 1.Zhenganxifeng Decoction has the characteristics of multi-system,multi-component and multi-target on treating Essential Hypertension,and the specific mechanism of Zhenganxifeng Decoction in treating hypertension has been preliminarily speculated and analyzed,these include the tumor pathway,the hepatitis B pathway,the Hypoxia-inducible Factors 1 pathway,the Phosphatidylinositol 3 kinase protein kinase pathway,and the Tumor necrosis factors pathway,The relevant targets are PI3 K,AKT1and IL6.2.Zhenganxifeng Decoction can reduce the blood pressure of the patients with primary hypertension(Grade 1)of Yin deficiency and Yang Hyperactivity Syndrome,improve the clinical symptoms,and is safe.It can be used to treat the patients with Essential Hypertension(grade 1)of Yin deficiency and Yang Hyperactivity Syndrome. |
