| Caesalpinia sinensis and Caesalpinia magnifoliolata,distributed mainly in Guangdong,Guangxi,Yunnan,Guizhou Provinces in China,belong to the Caesalpinia genus.The Caesalpinia genus is rich in cassane-type diterpenoids,which display extensive pharmacological activities,including anti-inflammatory,antioxidant,cytotoxic,antimalarial,and antibacterial activities.As part of a search for cassane-type diterpenoids from C.sinensis and C.magnifoliolata,a chemical investigation of the seeds of them were carried out,using silica gel column,Sephadex LH-20 column,preparative HPLC,and semiprepative HPLC.The structures of the isolated compounds were established by extensive spectroscopic analysis,and their absolute configurations were assigned by electronic circular dichroism calculations.In total,thirty-three compounds were isolated,including twenty-three cassane-type diterpenoids and three phenolic compounds from C.sinensis and three cassane-type diterpenoids and four other type compounds from C.magnifoliolata.Among them,the twenty new compounds are cassane-type diterpenoids,namely,(4S)-6β,12α,19-trihydroxy-cass-13(15)-en-16,12-olide(1),cass-13(15)-en-16,12-olide(2),12α-hydroxy-cass-13(15)-en-16,12-olide(3),12α-butoxy-6-oxo-cass-13(15)-en-16,12-olide(4),12α-ethoxy-6β-hydroxy-cass-13(15)-en-16,12-olide(5),6β-hydroxy-cass-13(14)-en-12-one(6),6β-hydroxy-cassan-12-one(7),6β-hydroxy-12-oxo-cassanic acid methyl ester(8),6β,7β-dihydroxy-cass-13(15)-en-16,12-olide(9),6β-formyloxy-cass-13(15)-en-16,12-olide(10),6β,11α,12α-trihydroxy-cass-13(15)-en-16,12-lactam(11),6β-hydroxy-12-oxocassanic acid ethyl ester(12),6β,12β-dihydroxy-cassanic acid(13),6β,12α,14β-trihydroxy-cass-13(15)-en-16,12-olide(14),6β,12α,14α-trihydroxy-cass-13(15)-en-16,12-olide(15),(4R)-6β,12α,18-trihydroxy-cass-13(15)-en-16,12-olide(16),2α,6β-dihydroxy-cass-13(15)-en-16,12-olide(17),2α,6β,12α-trihydroxy-cass-13(15)-en-16,12-olide(18),6β-hydroxy-12α-methoxy-cass-13(15)-en-16,12-lactam(19),6β,12α-dihydroxy-cass-13(15)-en-16,12-lactam(20).The thirteen known compounds are neocaesalpin WH(21),neocaesalpin WD(22),neocaesalpin WA(23),neocaesalpin WE(24),taepeenin L(25),(9Z)-glycerin-1-monooleate(26),(E)-9-octadecenoic acid(27),ethyl gallate(28),sappamide BC(29),botryosphaerin B(30),(+)-lariciresinol(31),(R)-O-methylmellein(32),4-hydroxymellein(33). |
