| Objective:To investigate the association of FOXM1 protein expression level in ovarian cancer cells nucleus or stroma with patient prognosis,and provide a basis for judging the prognosis of patients with ovarian cancer.Methods:The expression of FOXM1 protein in ovarian cancer cells nucleus or stroma was detected by tissue microarray technology and immunohistochemical method.Patients were divided into high and low expression groups based on the comprehensive score of immunohistochemical antibody staining intensity and positive rate,and the relationship between FOXM1 protein and prognosis of patients was analyzed in combination with clinical pathological and patient prognosis data.Results:1.A total of 154 sites were identified in the ovarian cancer tissue microarray with 133 actual effective sites,,of which FOXM1 protein is highly expressed in 26cases(19.5%)in tumor cell nucleus and 53 cases(39.8%)in stroma.2.There was no significant difference between FOXM1 protein expression level in ovarian cancer cell nucleus and age,FIGO stage,pathological type and stage of patients(P > 0.05),and there was no correlation with survival time of patients(P >0.05).3.There was a correlation between FOXM1 protein expression level in ovarian cancer interstitium and pathological classification of patients(P<0.05).The proportion of patients with high FOXM1 protein expression in serous carcinoma was higher than that in non-serous carcinoma.Survival analysis showed that higher FOXM1 protein expression level in tumor stroma was associated with lower progression free survival(PFS)(P<0.05).The overall survival rate(OS)of patients with high interstitial expression group was lower than that of patients with low stromal expression group,but OS had no statistical significance(P > 0.05).4.When the expression level of FOXM1 protein in the nucleus and stroma of ovarian cancer was analyzed together,the OS of FOXM1 protein decreased significantly in the group with high expression of both.5.In univariate analysis,FOXM1 protein expression level,age,FIGO stage,T stage,N stage,M stage,pathological grade and classification were all significant predictors of ovarian cancer survival.However,FIGO stage was only suggested as an independent prognostic factor in multivariate Cox regression analysis.Conclusion:1.When FOXM1 protein is highly expressed in both the nucleus and stroma of ovarian cancer,it has adverse effects on OS of patients;2.High FOXM1 protein expression in tumor stroma was associated with worse PFS in patients,but not with OS;3.The expression of FOXM1 protein in the nucleus and stroma of ovarian cancer is closely related to the prognosis of patients,and may be a new biomarker for predicting the prognosis of OC. |
