Objective:To explore the value of differential genes in clinical prognosis and the function of differential genes based on bioinformatics technology with oral squamous cell carcinoma in TCGA database.Methods:Based on the data set of TCGA who were diagnosed head and neck squamous cell carcinoma,all data were performed using R(http://www.r-projec.org,The R Foundation).Differential genes were screened under the condition of FDR < 0.05 and | 1og2 fc | > 3 for verification.According to the inclusion and exclusion criteria,Squamous cell carcinoma,from the Department of Oral and Maxillofacial Surgery,The First Affiliated Hospital of Shanxi Medical University and Shanxi Provincial People’s Hospital were recruited.SCC25 cells were transfected by sh RNA lentivirus interference technology.The effects of C1QL1 on apoptosis,migration,invasion and proliferation of oral squamous cell carcinoma were evaluated by flow cytometry test,CCK8,Transwell and scratch test.Results:We finally obtained 503 complete head and neck squamous cell carcinoma tissue samples and 44 normal tissue samples from TCGA database.A total of 646 differentially expressed genes,including 307 up-regulated genes and 339 down-regulated genes,were screened by bioinformatics analysis.Through literature review and GEPIA2 analysis,it was found that the high expression of C1QL1 gene was significantly correlated with the shortening of survival time of tumor patients.By WB,q PCR and immunohistochemistry,the expression level of C1QL1 in normal adjacent tissues was lower than that in oral squamous-cell carcinoma.Subsequently,we investigated the expression of C1QL1 in different oral squamous cell carcinoma cells in vitro and found that the expression of C1QL1 in SCC25 cells increased significantly Through sh RNA lentivirus interference with SCC25 cells,it was found that down-regulation of C1QL1 could inhibit the proliferation,invasion and migration of SCC25 cells.Conclusion:The expression of C1QL1 is high in oral squamous cell carcinoma tissues and inhibiting the expression of C1QL1 can inhibit the proliferation,invasion and migration of oral squamous cell carcinoma cells SCC25.OSCC could be a potential therapeutic target for OSCC. |
