The Role Of Borealin In The Proliferation,migration And Invasion Of Oral Squamous Cell Carcinoma And Its Molecular Mechanism

Objective: 1.To clarify the expression of Borealin in OSCC tissues and normal oral tissues,and to analyze the relationship between Borealin expression and clinicopathological factors of OSCC.2.To observe the effects of Borealin on the proliferation,migration and invasion of OSCC cells.3.To investigate the mechanism of Borealin on the proliferation,migration and invasion of OSCC cells.Methods: 1.Immunohistochemistry was used to detect the expression of Borealin in OSCC and chi-square test was applied to analyze the relationship between Borealin and clinicopathological factors in OSCC.2.The effects of Borealin on cell proliferation of OSCC cells were investigated by CCK-8 assay and plate clone formation assay.3.Flow cytometry was used to detect the effects of Borealin on cell apoptosis and cell cycle of OSCC cells.4.The effects of Borealin on invasion and migration of OSCC cells were examined by wound healing assay and transwell assay,respectively.5.Western Blot was used to detect the expression of Borealin on cycle-related proteins,apoptosis-related proteins,EMT-related proteins and relevant signaling proteins of OSCC cells.6.Transcriptional factors that regulate Borealin were predicted by bioinformatics.q RT-PCR and Western Blot were used to verify the regulation of transcription factors on Borealin.Results: 1.The expression of Borealin was up-regulated in OSCC tissues compared with normal oral tissues(P=0.0023),and its up-regulated expression was correlated with tumor size,lymph node metastasis and degree of differentiation(P<0.05),but not age and sex(P>0.05).2.Knockdown of Borealin inhibited OSCC cell proliferation,promoted OSCC cell apoptosis.When Borealin was knocked down in OSCC cells,the expression of Cyclin B1,Survivin,Aurora-B and Bcl-2 decreased,while the expression of Bax and Cleaved-Caspase3 was increased.3.Knockdown of Borealin inhibited OSCC cells migration and invasion.When Borealin was knocked down in OSCC cells,the expression of E-cadherin was increased,while the expression of N-cadherin and Snail was decreased.4.Knockdown of Borealin in OSCC cells inhibited the expression of p-Src and p-Akt,while the expression of Src and Akt did not change.5.Fox M1 was predicted to regulate the expression of Borealin by databases and websites.When Fox M1 was knocked down in OSCC cells,the expression of Borealin was decreased.Conclusions: 1.The expression of Borealin was up-regulated in OSCC,and its expression was closely related to clinical pathological factors.2.Borealin can promote cells proliferation and inhibit cells apoptosis.3.Borealin can inhibit the migration and invasion of OSCC cells,and Borealin expression has a significant correlation with the EMT process.4.Borealin may promote the proliferation,migration and invasion of OSCC through the Src/Akt signaling pathway.5.Fox M1 can promote the expression of Borealin in OSCC.
