Objective: On the basis of the cypress leaves washed party flavored preparation a conditioning shampoo of scalp psoriasis-Modified cypress leaves shampoo.The sensory index,physical and chemical index and skin irritation test of shampoo were investigated and evaluated,and applied to imiquimod induced psoriatic mouse model,to analyze its effect on psoriatic mouse model and related mechanism.To lay the foundation for the development of an appropriate dosage form,small side effects,accurate curative effect,simple use and easy to carry Chinese herbal compound shampoo,increasing the compliance and bringing good news for the majority of patients with scalp psoriasis.Methods:1.Orthogonal design was used to optimize the extraction process of raw liquid of Traditional Chinese medicine and the compounding process of surfactant,and the optimal design process was obtained,and the low,medium and three different concentrations of Modified cypress leaves shampoo were prepared.The sensory index of shampoo product was observed,and the heat and cold resistance test,p H value and foam height were measured.Finally,the product was applied to the skin of mice for skin irritation test.2.First,12 BALB/ C mice were set up as blank control group and model group,and modeling was carried out by referring to relevant literature.After modeling was successful,Then 60 BALB/ C mice were randomly divided into blank matrix group(CON group),model control group(IMQ group),Leyin shampoo group(LY group),Modified cypress leaves shampoo low concentration group(C-L group),Modified cypress leaves shampoo medium concentration group(C-M group)and Modified cypress leaves shampoo high concentration group(C-H group).After 8 days,the general condition and body weight trend of mice were observed,organ index was calculated,PASI score was performed on back skin lesions.The expression levels of IL-17 A m RNA and IL-23 m RNA in mouse skin lesions were detected by real-time quantitative PCR,and the protein expression levels of IL-17 A and IL-23 in mouse skin lesions were detected by enzyme-linked immunosorbent assay(ELISA).Results:1.In the orthogonal experimental design,the optimal extraction process of traditional Chinese medicine raw liquid was as follows: extraction time 1.5h,solid-liquid ratio 1:10,extraction times 3 times.The optimal dosage of surfactant in shampoo matrix:(AESA)12%,cocamide-diethanolamine(6501)3%,cocamide-propyl betaine(CAB-35)3%;The finished product of cypress leaves flavored shampoo has no foreign matter,light brown color and no pungent aroma.Through heat and cold resistance test,it was found that there was no precipitation,delamination and flocculation in shampoo products.The p H value of the dilution method was 6.8,and the foam height was 115 mm,which met the standard requirements.No erythema,edema and other reactions were found in the skin irritation tests of single administration and multiple administration.2.In the building experiment,compared with the blank control group,mice were smeared with imiquimod cream for 8 days for modeling,a typical erythema overlying white scales of psoriatic change,combining with HE staining showed obvious thickening of the epidermis keratin layer,parakeratosis companion hyperkeratosis,subcutaneous inflammatory cells infiltrating tips such as psoriasis sample model building success;In the formal administration experiment,after 8 days of administration,mice in each group were generally in good condition,except for CON group,body weight decreased to varying degrees.Compared with IMQ group,psorias-like lesions on the back were improved and PASI scores were decreased on the 8th day(P < 0.01).Compared with CON group,thymus index decreased in other groups on day 8,while spleen index increased,indicating that IMQ stimulation may cause thymus atrophy and spleen enlargement.The spleen index in LY,C-L and CH groups was lower than that in IMQ group(P< 0.01),indicating that inflammatory response was alleviated to a certain extent.Compared with CON group,the histopathological morphology of the other groups was changed.Compared with IMQ group,HE staining showed that the epidermal hyperplasia was reduced and the thickness was thinner in LY,C-L,C-M and C-H groups.Masson staining showed that the degree of fibrous tissue hyperplasia and collagen fiber deposition was reduced in CH,CM and LY groups;Real-time q PCR and ELISA results showed that compared with IMQ group,the expression levels of IL-17 A and IL-23 in LY,C-L,C-M and C-H groups were significantly decreased(P <0.01).Conclusion:1.Modified cypress leaves shampoo has good appearance,stable texture,weak p H,good foaming ability.It is safe to act on mouse skin without acute or chronic irritation.2.To some extent,the relative expression levels of IL-17 A and IL-23 were decreased,and the expression of inflammatory factors in psoriatic lesions was inhibited to block the activation of IL-23/IL-17-Th17 axis,so as to improve the severity of imiquimot-induced skin lesions in mice and reduce the proliferation of keratinocytes and fibroids in epidermis.The spleen index of mice was decreased,indicating that the Chinese herbal compound shampoo could regulate psoriasis of scalp.
