| Objective:This topic was based on ultra performance liquid chromato g-raphy tandem quadrupole time of flight mass spectrometry(UPLC-MS/MS Q-TOF)technology,screened the differential metabolites in the urine samples of adult female patients diagnosed as Kashin-Beck disease(KBD)by non targete dmetabonomics,and explored the metabolic pathways and their biological funct i-ons of these metabolites involved,in order to provide a certain clue to the p at-hogenesis of KBDMethod:According to the results of monitoring and field investigation of KBD regions in Gui De and Xing Hai countries in Hainan Tibet Autonomous Prefecture,Qinghai Province,30 adult female patients over 20 years old were selected as case group,30 female located in local district as control group.10 ml of morning urine after fasting was collected from the study population.The type,ingredient,content and changes of low molecular weight metabolites in u-rine were detected with UPLC-MS/MS Q-TOF technology,then,combining multi-statistical analysts and the least squares method differentiation analysis(PLS-DA),with the HMDB database and METLIN database alignment to screen dif-ferentmetabolites.Result:12,793 metabolites from the urine samples were detected,and85 different metabolites were screened among of them,7 metabolites were upregulatedand 78 metabolites were down-regulated.The different metabolites wer-e filteredby HMDB database and related literature search,and compared with the secondary mass spectrogram.Finally,14 different metabolites were identifie-d,there are four different metabolites in anionic mode,including aconitic acid,anti-2-crotonic acid,Acetyl-N-formyl-5-methoxykynurenamine and citrulline,whi-le,there were ten different metabolites in cationic mode,includingα-Keto-glut-aramicacid,uracil,tryptophanol,N-acetyl-5-hydroxytryptamine,4,6-dihydroxyqui-noline,3-hydroxybutyric acid,pyridoxine 5’-phosphate,5’-methylthioadenosine,β-D-glucose and glucosamine,which were involved tryptophan,vitamin and oth-er metabolic pathways.Conclusion: There are existing abnormal changes in urine of female KBD disease patients,14 kinds of down-regulated metabolites might be related with oxidative damage,inflammatory response and involve in tryptophan metab-olism,abnormal vitamin metabolism and other metabolic damages. |
