Objective To investigate the relationship between lncRNA TUG1 and clinicopathological characteristics of patients with colorectal cancer(CRC),and the effect of lncRNA TUG1 on chemosensitivity of colorectal cancer cells.Methods The clinical data of patients with CRC in Jiaxing First Hospital from September 2020 to June 2022 were collected to analyze the relating factors of metastatic and recurrence in CRC.RT-qPCR were used to detect TUG1 expression in tissues and analyzed the relationship between TUG1 and clinicopathological characteristics of CRC.Cell experiments were further carried out to analyze the 5-fluorouracil chemosensitivity mediated by TUG1 in colorectal cancer cells.Results LncRNA TUG1 was significantly up-regulated in CRC tumor samples compared with the surgical margins(P<0.001).The clinical data analysis showed that the progression of tumor metastasis after postoperative chemotherapy of CRC was correlated with the degree of differentiation,the level of lncRNA TUG1 expression(P<0.05).The expression of lncRNA TUG1 was correlated with periintestinal regional lymph node metastasis and vascular cancer thrombus of colorectal cancer(P<0.05),but not with age,sex,body mass index,tumor location,tumor size,pathological type,degree of differentiation,depth of invasion,nerve invasion,P53,Ki67 and tumor budding(P>0.05).Multivariate analysis revealed that lymph node metastasis in the periintestinal region was an independent risk factor for increased lncRNA TUG1 expression(OR=2.349,95%CI:1.003-5.501,P=0.049).In cell experiments,compared with the control group,the cell survival rate and IC50 in si-TUG1 group were decreased and the apoptosis rate was increased after treatment with 5-fluorouracil(P<0.05).The expression of E-cadherin in si-TUG1 group was higher than that in the control group(P<0.05).Conclusion The expression of lncRNA TUG1 in CRC tumor samples is increased.The metastasis and recurrence of CRC are related to the degree of tumor differentiation and the expression level of lncRNA TUG1.The expression of lncRNA TUG1 is related to the periintestinal lymph node metastasis and vascular tumor thrombus in CRC,and lymph node metastasis in the periintestinal region is an independent risk factor for the expression level of lncRNA TUG1.Knockdown of lncRNA TUG1 causes an increased sensitivity of CRC cells to 5-fluorouracil,its mechanism may be related to the up-regulation of E-cadherin expression.LncRNA TUG1 may become a new therapeutic target for reversing 5-fluorouracil resistance in CRC,and provide new ideas for clinical treatment and basic research.
