Objective:The study was conducted to determine the expression levels of Lys-Asp-Glu-Leu Receptor 2(KDELR2)m RNA and protein in glioma,further to explore the relationship betweenlevelchanges of KDELR2 with glioma clinical indications and patient prognosis。Methods:1.A tissue microarray(HBra G180Su01)containing standard clinical and pathological data was used.Tissue microarray sections prepared as above were used for immunohistochemistry experiments by Streptomycin-Peroxide method;All slides were scanned using Image Scope and analyzed with Image J software for Immunoreactive Score(IRS).2.All data were processed by R(version 4.0.4),and we applied Chi-square test to determine the association of KDELR2 protein expression with each of the patients’ clinical pathological parameters.Survival curves were generated by using the Kaplan-Meier method,and differences in survival were evaluated with the log-rank test.Patient overall-survival was modeled,P < 0.05 was considered statistically significant.Multivariate Cox regression was used to confirm the relationship between levelchanges of KDELR2 with glioma clinical indications and patient prognosis.3.To provide a wider dynamic change of KDELR2 in pan-cancer analysis,we analyzed pan-cancer cases in the Sanger Box,including gene expression,prognostic analysis,and immunityinfiltration.Finally,we further investigate the role of KDELR2 m RNA in the relationship between gene expression and prognostic analysis.Results:1.KDELR2 is mainly membranous or cytoplasmic staining,the percentage of positive rate was 98.18 %.2.KDELR2 expression was significantly correlated with tumor grades,age,recurrent status,survival status,PDL1 expression(P < 0.05),and no significant associations with sex,Ki67,EGFR(P>0.05).165 samples are analyzed by Univariate Kaplan-Meier analyses showed that age,tumor pathological grade,KDELR2 were significantly associated with overall survival(P < 0.05).in addition,age,tumor pathological grade,KDELR2,EGFR were significantly associated with Progression Free Survival(P<0.05).The median OS and PFS were elevated in the high expression groups of KDELR2 compared to low expression counterparts.A multivariate Cox regression analysis showed that tumor pathological grade,KDELR2 were identified as independent predictors of glioma.In the higher expression group,the risk of mortality was 2.45-fold change,and recurrence risk was 2.34 fold change than lower group.3.In pan-cancer analysis,expression of KDELR2 was much stronger in tumor tissues compared with paired paracarcinoma tissues,wassignificantly associated with the prognosis of a variety of tumors,and was related to prominently with many kinds of tumor and immune cells infiltration.Conclusions:1.KDELR2 expression level was significantly associated with grade,age,recurrent status,survival status,PDL1 expression in glioma.2.High KDELR2 expression was associated with poor prognosis,and an important factor of glioma relapse and progression,the higher KDELR2 expression level was associated with poorer outcomes.Therefore,KDELR2 may serves as a valuable biomarker and attractive therapeutic targets for glioma treatment.3.KDELR2 wassignificantly associated with in immune infiltration.
