| Objective:To explore the mechanism of "softening tendons and setting bones" massage therapy in the treatment of knee osteoarthritis by experimental study on the influence of apoptosis of articular chondrocytes in rabbit model of knee osteoarthritis,in order to provide theoretical basis for the clinical application of "softening tendons and setting bones" massage therapy.Methods:A total of 24 clean male New Zealand white rabbits were selected,7 were randomly selected as blank group,and the remaining 17 were modeled uniformly.Methods The injection of papain into knee cavity was used.After the modeling was completed,3 model rabbits were killed randomly and the articular cartilage was observed by naked eye.After successful modeling,the remaining 14 rats were randomly divided into two groups:model control group and intervention group,with 7 rats in each group.The intervention group was treated with "soft tendons and bone-setting" massage,once every other day,a total of 4 weeks;The model control group and the blank group did the same grasping and fixation atthe same frequency-and time as the intervention group,without any other intervention.The modified Lcqucsnc MG evaluation method was used to conduct behavioral evaluation before and after intervention.After treatment,rabbits in each group were sacrificed,and knee cartilage tissues were collected.HE staining and Mankin’s score were used to evaluate the structural and morphological changes of cartilage tissue.The inflammatory cytokines IL-1β and TNF-α were detected by enzyme-linked immunosorbent assay(Elisa).The contents of Bcl-2,Bax and Caspase-3 in cartilage were determined by Western Blot.Results:According to behavioral evaluation of modified Lcqucsnc MG evaluation method,the left posterior knee joint of model rabbits showed obvious pain,joint enlargement accompanied with decreased range of motion,and lameness during walking.There was no abnormality in the left hind limb of blank group before and after modeling.The intervention group was treated on the basis of successful modeling,pain was relieved,gait and joint.movement were improved.Mankin’ s score of cartilage tissue was used to evaluate the morphology of chondrocytes in blank group,and the score was 0.In model control group,the cartilage surface was damaged obviously,there were cracks,the number of cells was reduced,the tide line was broken,and vascular invasion could be seen.The cell morphology and structure of the intervention group were significantly improved compared with the model control group.The levels of IL-1 β and TNF-αin rabbit arterial blood were the lowest in blank group,and the highest in model control group.After intervention,the content.in the intervention group decreased significantly compared with the model control group,and there were significant differences between the groups(P<0.05).Western Blot analysis showed that the expression level of Bcl-2 protein was the highest in blank group,the lowest in model control group,and the expression level in intervention group was higher than that in model control group after intervention.The expression levels of Bax and Caspase-3 protein in blank group were the lowest,while the expression levels in model control group were the highest.After intervention,the expression levels in intervention group were significantly decreased compared with model control group,and there were significant differences between groups(P<0.05).Conclusion:"Softening tendons and boning" massage can effectively reduce the expression of Bax and Caspase-3,improve the expression of Bcl-2,inhibit the apoptosis of chondrocytes,promote inflammatory absorption,reduce the degree of cartilage destruction,and play a role in protecting cartilage. |
