Objective To explore the expression of Toll-like receptor 2(TLR2)on basophils in peripheral blood of patients with allergic rhinitis(AR),allergic asthma(AA),allergic rhinitis combined with asthma(ARA)and healthy controls(HC)before and after allergen stimulation;and in blood of sensitized mouse models,so as to explore the potential role of TLR2 in the above allergic diseases,and provide the date for clinical trials.Methods In this study,a total of 15 HC subjects,42 AR patients,27 AA patients,and 19 ARA patients were collected with 3 mL of peripheral blood.The flow cytometry was used to detect the changes expression of TLR2 on peripheral blood basophils before and after the stimulation with House dust mite allergen extract(HDME),Artemisia sieversiana wild allergen extract(ASWE),and Platanus pollen allergen extract(PPE).Meanwhile,AR,AA mouse models were established to explore the expression of TLR2 on basophils in blood and mast cells in lung tissue of mice.Results1.In the three kinds of basophil marker: CCR3~+(CC chemokine receptor 3),CD123~+HLA-DR~-(human leucocyte antigen DR),CD123~+HLA-DR~-CCR3~+ granulocytes and PBMC(peripheral blood mononuclear cell),the percentage of TLR2~+ cells on blood of patients with three diseases was lower than that of healthy controls.The MFI(mean fluorescence intensity)of TLR2 on CCR3~+,CD123~+HLA-DR~-,CD123~+HLA-DR~-CCR3~+ granulocytes and PBMC also decreased.After stimulation with different allergens(HDME,ASWE,PPE),the expression of TLR2 in patient’s peripheral blood also decreased to different degrees.2.In AR and AA mouse models,the expression of TLR2 on blood basophils and Mast cells of lung tissue with WT(wild type)and FcεRⅠ KO(Fc ε receptor Ⅰ knockout)mice were decreased to varying degrees compared with normal saline(NS)-NS group,and the percentage of TLR2~+ cells in WT mice was lower than that of FcεRⅠKO mice.Conclusions1.The expression of TLR2 on basophils in patients of three diseases were decreased;when stimulated by different allergens,the percentage of TLR2~+ cells on basophils also decreased to different degrees.Suggesting that basophils may participate in the occurrence of AR,AA through TLR2.2.The percentage of TLR2 on basophils and Mast Cell in sensitized mouse model also decreased,which was consistent with human results.Comparison of two kinds of mice shows that the deletion of FcεRⅠwill affect the expression of TLR2.
