Purpose:In this study,medical records of CAG patients in Professor Yao Wei’s outpatients from June 2018 to June 2022 were taken as the research object,relevant information in medical records were analyzed,and the academic thoughts and core herbs of Professor Yao Wei in the treatment of CAG were summarized.Network pharmacology and molecular docking of core herbs were studied to explore the mechanism for providing new diagnosis,treatment ideas and research direction for clinical treatment of CAG.Method:1.Medication rules and academic thoughts of Professor Yao Wei in the treatment of CAGThe sorted medical record information was input into the TCM inheritance assistance platform for analysis.The gender and age of the patients,the TCM syndrome types of CAG,the four Qi,five flavors,meridian conversion,high-frequency herbs and core herbs were summarized in the treatment,and the curative effect of Professor Yao Wei’s treatment of CAG was analyzed by comparing the medical records of the first diagnosis and the return diagnosis one month later,and summarized Professor Yao Wei’s academic thoughts on CAG treatment.2.Network pharmacology and molecular docking of core herbsNetwork pharmacological research was carried out on Professor Yao Wei’s core herbs in the treatment of CAG(Zhi Shi,Hou Po,Dang Shen,Bai Zhu,Fu Ling,Zhi Gan Cao,Chen Pi,Ban Xia,Mu Xiang,Fo Shou,Hai Piao Xiao,Zhe Bei Mu,Pao Jiang),in order to explore the potential mechanism of action of core herbs by analyzing their related active ingredients,target proteins and related signaling pathways.The binding ability of the selected active ingredients and target proteins was verified by molecular docking studies.Results:1.Medication rules and academic thoughts of Professor Yao Wei in the treatment of CAGA total of 311 patients with CAG were collected and 283 were included,including120 males and 163 females.The number of people aged 50-59 and 60-69 was relatively large,accounting for more than 50%.In the analysis of traditional Chinese medicine syndrome,it was found that spleen-stomach weakness and liver-stomach Qi stagnation were the most common syndromes in CAG patients,accounting for more than 50%.According to the analysis of the using of traditional Chinese medicine for CAG patients in medical records,in the four Qi of traditional Chinese medicine,warm and peaceful herbs were the main herbs,and warm and peaceful herbs accounted for more than 79%of all the traditional Chinese medicine.Bitter,spicy and sweet were significantly more than other flavors among the five flavors of traditional Chinese medicine,accounting for more than 85% of the total.The spleen,stomach,lung and liver channels account for more than 68% of the total channels.According to the four Qi,five taste and channel channels of traditional Chinese medicine,Professor Yao Wei’s rule of dispensing prescription in clinical treatment of CAG patients can be summarized,that is,the main herbs using should be warm and peaceful,and the using of cold and heat herbs should be cautious to prevent the injury.Using bitter dryness herbs and sweet moistening herbs to keep a balance;Paying attention to regulate the machine of Qi;Paying attention to the spleen and stomach,both liver and lung.Through the screening of herbs in the medical records,13 core herbs were obtained:Zhi Shi,Hou Po,Dang Shen,Bai Zhu,Fu Ling,Zhi Gan Cao,Chen Pi,Ban Xia,Mu Xiang,Fo Shou,Hai Piao Xiao,Zhe Bei Mu and Pao Jiang.Among them,Zhi Shi and Hou Po can eliminate ruffian full;Dang Shen,Bai Zhu,Fu Ling,Zhi Gan Cao,Chen Pi,Ban Xia combined for Liu Jun Zi Tang,can be replenishing Qi to invigorate the spleen;Mu Xiang and Fo Shou can be soothing liver and clearing heat,promoting Qi to activate blood;Hai Piao Xiao and Zhe Bei Mu can treat epigastric pain and vomiting acid water;Pao Jiang can mild stomach.At the same time,the selected core herbs also meet the herb characteristics of Professor Yao Wei in the treatment of CAG.Bitter herbs such as Zhi Shi,Chen Pi and Fo Shou can relieve dampness.Zhi Gan Cao,Dang Shen and other sweet herbs with Bai Zhu have the meaning of nourishing;Ban Xia,Mu Xiang and other spicy herbs can regulate Qi.It reflects Professor Yao Wei’s academic thought of focusing on regulating Qi,taking into account both dryness and wetness,and taking a flat period in the treatment of CAG.2.Network pharmacology and molecular docking of core herbsAfter screening the core herbs,203 active ingredients and 284 target proteins of the core herbs were obtained,among which 97 overlaps with CAG disease target proteins,and 97 overlaps were involved in 87 functional items and 160 signaling pathways,among which the chemical-carcinogenic ROS signaling pathway ranked higher,and CAG has the most closely related to ROS.Quercetin,luteolin and baicalin in the composition of the core herbs act on AKT1 and VEGFA target proteins,and participate in the chemical carcinogenic ROS signaling pathway through PI3K-Akt and VEGF signaling pathway,playing antioxidant and anti-inflammatory effects.By molecular docking studies,quercetin,luteolin and baicalin can form stable chemical interaction with AKT1 and VEGFA.Conclusion:1.By analyzing the outpatient medical records of CAG patients treated by Professor Yao Wei,this study summarized the core herbs used by Professor Yao Wei in the treatment of CAG,namely,the core herbs were Zhi Shi,Hou Po,Dang Shen,Bai Zhu,Fu Ling,Zhi Gan Cao,Chen Pi,Ban Xia,Mu Xiang,Fo Shou,Hai Piao Xiao,Zhe Bei Mu and Pao Jiang.The core idea was that the etiology of CAG lies in the mixture of dryness and wetness,and the clinical herbs should be warm and peace;The using of cold and heat herbs should be cautious to prevent the injury;Using bitter dryness herbs and sweet moistening herbs to keep a balance;Paying attention to regulating the machine of Qi;Paying attention to the spleen and stomach,both liver and lung.2.After the analysis of these core herbs through network pharmacology and molecular docking,it is speculated that the mechanism of these core herbs is mainly through the combination that quercetin,luteolin and baicalin act on AKT1 and VEGFA target proteins,and participate in the chemical carcinogenic ROS signaling pathway through PI3K-Akt and VEGF signaling pathway,playing antioxidant and anti-inflammatory effects,so as to achieve the therapeutic effect on CAG.
