Objective:With three Alpinia Roxb components of flavonoids as research object,with copy of rats gastric ulcer model of cold syndrome,on three Alpinia Roxb components of flavonoids medicinal properties taste,rats by observing its systemic function index determination,energy metabolism,endocrine system,sympathetic nerve-adrenal system,the influence of cyclic nucleotide system,sum up out the common medicinal properties of the foundation,analyzes the possible intrinsic relations,based on related medicinal effect material in basic research to provide experimental basis.Methods:(1)The flavonoids of 3 traditional Chinese medicines were extracted by ethanol reflux extraction;The flavonoids were purified and enriched by alkaline solution and acid precipitation method.(2)Ninety SD male rats were selected,and 10 were randomly selected as blank control group.The rest of the rats were treated with ice acetic acid and 4℃ice anemonium decoction to replicate the rat model of cold syndrome of gastric ulcer.After successful model replication,the rats were divided into model group,positive ginger group,Alpinia officinarum Hance,Alpinia galangal L.,Alpinia galanga Willd of three flavones,high and low dose groups.The model group and blank group were given 0.5%Twain 80 aqueous solution,and the other groups were given corresponding drugs by gavage.The body temperature indexes(head,neck,left armpit,right armpit,left groin,right groin,chest,upper abdomen and middle abdomen)of rats with cold syndrome of gastric ulcer were measured by infrared thermal scanner.The formation of gastric ulcer was observed under magnifying glass and the gastric ulcer index was calculated.The contents of Na+-K+-ATPase,Ca2+-Mg2+-ATPase,SDH and liver glycogen in rats were measured with the kit.The contents of TSH,T3,T4,ACTH,D-β-H,FFA,LIPD and LIPC in serum and the contents of AC,NEPP2,c AMP,c GMP,Na+-K+-ATPase and Ca2+-Mg2+-ATPase in gastric tissues were determined by ELISA.SPSS 22.0 was used for statistical analysis,and P<0.05 was considered statistically significant.Results:(1)Effect on functional activity of ratsCompared with the blank group,the ulcer area of the model rats with cold syndrome of gastric ulcer increased significantly(P<0.01),and the temperature of upper abdomen,middle abdomen and right groin decreased significantly(P<0.01 or P<0.05).Compared with the model group,the gastric ulcer index of rats in the high and low dose groups of Alpinia officinarum Hance,Alpinia galangal L.,and Alpinia galanga Willd was significantly decreased,the difference was statistically significant(P<0.01 or P<0.05).The temperature of upper and middle abdomen of rats in the high and low dose groups of Alpinia officinarum Hance and Alpinia galangal L,the high dose groups of Alpinia galanga Willd could be significantly increased(P<0.01 or P<0.05).(2)Effect on energy metabolism of ratsCompared with blank group,the activities of Na+-K+-ATP,Ca2+-Mg2+-ATPase and SDH in liver tissue and stomach tissue of rats with cold syndrome of gastric ulcer were significantly decreased,and liver glycogen level was significantly increased(P<0.01 or P<0.05).Compared with model group,the levels of Na+-K+-ATPase and SDH in liver tissue were significantly increased in Alpinia officinarum Hance,Alpinia galangal L.,Alpinia galanga Willd high and low dose groups(P<0.01 or P<0.05),and the levels of liver glycogen in liver tissue of rats were significantly decreased(P<0.05).The Ca2+-Mg2+-ATPase level in liver tissue was significantly increased in Alpinia officinarum Hance high-dose groups,Alpinia galangal L.,Alpinia galanga Willd high and low dose groups(P<0.01),and the Na+-K+-ATPase content in stomach tissue of rats was significantly increased in Alpinia galangal L high-dose groups,and Alpinia officinarum Hance,Alpinia galanga Willd high and low dose groups(P<0.01 or P<0.05),The contents of Ca2+-Mg2+-ATPase in gastric tissues of rats were significantly increased in Alpinia officinarum Hance high-dose group,Alpinia galangal L.,Alpinia galanga Willd high and low dose groups(P<0.01 or P<0.05).(3)The effect on the cyclic nucleotide systemCompared with blank group,content of AC,c AMP and c AMP/c GMP in gastric ulcer cold syndrome model group was significantly decreased,and NEPP2 content was significantly increased(P<0.01).Compared with model group,the content of AC in the gastric tissue of rats in the low dose group,Alpinia officinarum Hance and Alpinia galangal L.high-dose groups was significantly increased(P<0.01 or P<0.05);The content of NEPP2 in gastric tissue of rats in high and low dose groups was significantly decreased,the gastric c AMP content and c AMP/c GMP ratio of Alpinia officinarum Hance,Alpinia galangal L.,Alpinia galanga Willd high and low dose groups were significantly increased(P<0.01 or P<0.05).(4)The influence on metabolic axisCompared with blank group,serum TSH,T3 and T4 contents in gastric ulcer cold syndrome model group were significantly decreased(P<0.01).Compared with model group,serum TSH content in Alpinia officinarum Hance high-dose group and Alpinia galanga Willd high and low dose groups was significantly increased(P<0.01 or P<0.05);Alpinia officinarum Hance high dose group,Alpinia galangal L.,Alpinia galanga Willd high and low dose group of rats serum T3 content significantly increased(P<0.01 or P<0.05);The serum T4 content of Alpinia officinarum Hance and Alpinia galanga Willd high-dose groups was significantly increased(P<0.01 or P<0.05).(5)The effect on lipid metabolismCompared with the blank group,the serum levels of FFA,LIPD and LIPC in the model group were significantly decreased(P<0.01).Compared with model group,the serum FFA level in Alpinia officinarum Hance,Alpinia galangal L.,Alpinia galanga Willd high and low dose groups was significantly increased(P<0.01 or P<0.05).The levels of serum LIPD and LIPC in Alpinia galanga Willd high and low groups,Alpinia officinarum Hance and Alpinia galangal L high groups were significantly increased(P<0.01 or P<0.05).(6)The influence on the sympathetic nerve-adrenal systemCompared with blank group,serum ACTH and D-β-H contents in gastric ulcer cold syndrome model group were significantly decreased(P<0.01).Compared with model group,the contents of ACTH and D-β-H in serum of Alpinia officinarum Hance,Alpinia galangal L.,Alpinia galanga Willd in high and low dose groups were significantly increased(P<0.01 or P<0.05).Conclusion:(1)Three Alpinia Roxb components of flavonoids have the effects of increasing temperature,regulating energy substances,glycogen and lipid metabolism,regulating cyclic nucleotide system,and promoting the functional activities of thyroid system and sympatho adrenal system in cold ulcerated rats with gastric ulcer,mechanism of action of releasing cold may related to adjust these system related indexes.(2)Research shows that:indicating that flavonoids from Alpinia officinarum Hance,Alpinia galangal L.,Alpinia galanga Willd are as warm as their whole ingredients Heat resistance;as well as the similarity of drug resistance in the same genus of traditional Chinese medicine with close relatives;it was that one of three Alpinia Roxb components of flavonoids are the chemical basis of its embodied warm resistance.
