Objective: Through data mining technology,the medication rules of modern Chinese medicine for treating sterility of polycystic ovary syndrome(PCOS)with liver depression and kidney deficiency were studied.On this basis,the core prescriptions of modern doctors obtained from data mining were analyzed by network pharmacology,and new prescriptions for treating the disease were deduced.To predict the mechanism of action of core prescription in treating this disease.Methods: Literature related to the treatment of infertility caused by polycystic ovary syndrome with liver depression and kidney deficiency was collected from CNKI,Wanfang and VIP databases.Effective TCM prescriptions were obtained through inclusion criteria and exclusion criteria,which were unified sorted out and input into the TCM inheritance assistance platform(V2.50 version),and the data were statistically analyzed.The obtained core prescription drugs were put into the TCMSP database one by one,and the oral bioavailability was set to be ≥30% and drug-like drugs ≥0.18 for screening,so as to obtain the effective ingredients of traditional Chinese medicine.Genes associated with the disease were searched in Gene Cards and OMIM databases.At the same time,Uni Prot database was used to correct the gene names corresponding to the targets,and Cytoscape software(3.8.0)was used to construct the effective compound-target network-protein interaction network of the core prescription,and the core compounds and targets were screened out.Finally,DAVID database was used to conduct GO functional enrichment analysis and KEGG pathway enrichment analysis to study the possible mechanism of action.Results:(1)Data mining: A total of 98 prescriptions and 160 TCM were included after screening,and the top 15 TCM with the frequency of drug use included: Achyranthes bidentum,Radix Rehmanniae radix,cuscuta cuscuta,Radix paeoniae paeoniae,angelica sinensis,Cornus officinalis,bupleurum bupleurum,Chinese yam,Chinese herb Herb,Poria cocos,Salvia miltiorrhiza,Rhizoma atractylodes,wolfberry,Epimedium,Ligusticum ligusticum,licorice,and sequestrum(among them,sequestrum ligusticum,ligusticum ligusticum,and licorice 3 TCM use frequency is the same,ranked 15th).The four qi in Chinese medicine are mainly warm,flat and cold.The five tastes are sweet,bitter and sweet.The top three are liver,kidney and spleen channels.Association rules were used to dig prescription formulation rules and core prescriptions.By setting different levels of support and confidence,combined with the analysis of the excavated literature and the instructor’s experience in the clinical treatment of this disease,7 pairs of high-frequency drugs were obtained when setting association rules with 40 support degrees and 0.7confidence degrees,including 6 traditional Chinese medicines: Bupleurum bupleurum,Rehmanniae rehmannium,Radix anthus sinensis,semen cuscuta and Paeoniae Radix,the core prescription of infertility caused by liver stagnation and kidney deficiency polycystic ovary syndrome was obtained.A total of 11 new formula combinations were obtained through cluster analysis,as shown in the text.(2)Network pharmacology: The core prescriptions and diseases obtained through data mining analysis will be constructed for network pharmacology analysis.(1)50 effective ingredients in the core prescription for intervention of the disease were obtained,and 109 targets were obtained by the intersection of drugs and disease.(2)The protein interaction network was analyzed,and the core targets in the interaction network were estimated to be TP53,MAPK1,MAPK14,AKT1,EGFR,HSP90AA1,ESR1,STAT1,etc.,according to the degree value.(3)GO enrichment analysis was performed on the intersection targets,and 688 biological process items,70 cell composition items and 126 molecular function items were obtained.Biological processes are mainly concentrated in the positive and negative regulation of apoptotic process,cell response to drugs,response to foreign biological stimuli,positive regulation of gene expression,positive regulation of DNA transcription,response to hypoxia and so on.Cell composition is mainly concentrated in macromolecular complex and extracellular void.The molecular functions are mainly concentrated in transcription factor binding,enzyme binding,identical protein binding,protein kinase activity,protein kinase binding,protein homodimer activity,etc.(4)KEGG pathway enrichment analysis,set P < 0.01,160 signaling pathways were obtained.It mainly includes m TOR signaling pathway,AMPK signaling pathway,BCR signaling pathway,c AMP signaling pathway,cytosolic DNA sensing signaling pathway,Insulin signaling pathway,c GMP-PKG signaling pathway,etc.Conclusion(s):(1)Modern TCM doctors treat infertility caused by polycystic ovary syndrome of liver depression and kidney deficiency.According to this syndrome type,they basically use the method of soothing liver and relieving depression,tonifying kidney and filling essence to treat the disease.Comprehensive clinical experience of various doctors in recent years,through data mining,the core prescription includes cooked Rehmannia radix,Radix angica sinensis,Radix paeoniae paeoniae,dodder seed,bupleurum,Radix Xiangfu,this 6 traditional Chinese medicine.As for the new prescriptions obtained by clustering,there are great differences in drug classification composition among new prescriptions,and the efficacy of new prescriptions needs to be verified clinically.(2)Through the network pharmacological analysis,it is concluded that most of the compounds,targets and related signaling pathways in the core position of the regulatory network have been closely related to infertility caused by polycystic ovary syndrome,which has been confirmed by a large number of clinical practices or experiments.In addition,there are some nodes in the core position of the network,which are rarely studied on infertility caused by PCOS.It needs to be further studied in the future.In conclusion,the etiology and pathogenesis of infertility caused by polycystic ovary syndrome are complex,but the existing studies show that traditional Chinese medicine can play a role in the treatment of infertility caused by polycystic ovary syndrome in multiple ways and multiple targets.The quercetin,phytosterol,luteolin,kaempferol and other core compounds rich in this core prescription can regulate AMPK,Insulin,BCR,c GMP-PKG,m TOR and other signaling pathways.Change the expression of HSP90AA1,TP53,AKT1,MAPK14,MAPK1,ESR1,EGFR,TNF and other proteins,achieve anti-apoptosis,anti-oxidative stress,anti-inflammatory and other effects,play a certain role in delaying the course of infertility caused by polycystic ovary syndrome,protect the ovarian function of PCOS patients of childbearing age.Improve their fertility as much as possible.
